黄芪皂苷的分离纯化及其与PAI-1的相互作用研究

摘  要:分离黄芪饮片化学成分并研究黄芪皂苷与纤溶酶原激活物抑制物(PAI-1)的相互作用关系。采用D101大孔树脂柱、正向硅胶、Sephadex LH-20、MCI、ODS等各种分离填料进行皂苷分离纯化,用紫外-可见光谱法研究药物分子与PAI-1的相互作用。并采用双倒数法进行实验数据处理分析。结果表明:PAI-1与黄芪皂苷Ⅰ、Ⅱ、Ⅳ的结合常数均大于与人参皂苷Rg1的结合常数K值,AstragalosideⅡ及AstragalosideⅣ具有潜在的较好抗血栓活性。To isolate bioactive constituents from astragalus and study the interaction of saponins with Human Plas minogen activator inhibitor-1. Isolation and purification were performed through D101 macroporous resin, silica gel column chromatograph, Sephadex LH-20, MCI column chromatograph, and reversed-phase ODS column. The vitro bioactive study through investigating the binding of saponins with PAI-1 by UV-vis spectroscopy. And the Double Reciprocal equation was applied to analyse the experimental data. The Astragaloside I , Astragaloside 11 and Astragaloside 1V have higher binding constant with PAI-1 than Ginsenoside-Rgl. Astragaloside 11 and Astragaloside IV potentially have good antithrombotic activity.
