摘  要:目的探讨幼年大鼠感染性脑损伤时外周血中高迁移率族蛋白B1(High mobility group protein B1,HMGB1)的表达规律及丙酮酸乙酯(Ethyl pyruvate,EP)对其的保护作用。方法 150只1月龄SD大鼠随机分为对照组(NS组,50例),颈外动脉注射生理盐水;感染性脑损伤组(LPS组,50例),颈外动脉注射LPS;EP治疗组(EP组,50例),颈外动脉注射LPS后即刻腹腔注射EP,于注射药物后6、12、24、48、72 h取外周静脉血,采用酶联免疫吸附试验分析HMGB1、神经元烯醇化酶(Neuron specific enolase,NSE)水平。结果 LPS组外周血HMGB1、NSE水平显著增高,各时间点与NS组比较差异有统计学意义。EP干预后外周血HMGB1及NSE水平均显著下降(P均<0.05),LPS组及EP组HMGB1与NSE水平均呈正相关。结论 HMGB1可能作为晚期炎症因子参与感染性脑损伤的发生发展过程,外周血HMGB1的检测可较好地反映感染性脑损伤时HMGB1的表达规律;丙酮酸乙酯可能通过抑制HMGB1的表达而对感染性脑损伤具有保护作用。Objective To investigate the serum levels of high mobility group box 1(HMGB1) in the infant rats of infectious brain injury.Methods One hundred and fifty normal healthy 1-month-old Spragne-Dawley(SD) rats were randomly divided into three groups: LPS group(n=50),in which rat was injected LPS via external carotid; NS group(n=50),injected NS via external carotid; EP group(n=50) after injecting LPS,immediately administrated EP intraperitoneally.Venous blood samples were collected at 6,12,24,48 and 72 h following drug injection.Their serum levels of HMGB1 and NSE were measured by ELISA.Results After LPS was given,serum levels of HMGB1 and NSE started to rise significantly.Compared with NS groups at each time point,the difference was significant(P0.05).In EP group,the levels of HMGB1 and NSE were decreased,the difference was significant(P0.05).Positivecorrelation were still gotten among HMGB1 and NSE in LPS and EP groups.Conclusion HMGB1 seems to act as a late mediator in the process of infectious brain injury,detection of serum HMGB1 can better reflect its expression in infectious brain injury.EP has neuroprotective effect on infectious brain injury,which maybe relevant to decreasing the levels of HMGB1 and suppressing inflammation action.
