摘 要:目的通过建立房颤大鼠模型,观察β2肾上腺素能受体(β2-AR)mRNA的表达水平以及青蒿提取物对其表达的影响。方法大鼠随机分为对照组、模型组、青蒿组、卡托普利组。利用舌下静脉注射乙酯胆碱和氯化钙的方法建立房颤大鼠模型,使用青蒿提取物进行干预,以卡托普利作为对照组。实验前2周青蒿组以1.25 g生药/kg灌胃,卡托普利组以3.25 mg/kg灌胃,对照组及模型组均以等体积生理盐水灌胃。PCR及Western-blot检测β2-AR蛋白及其mRNA表达。结果与对照组相比,模型组大鼠心肌β2-AR mRNA的表达显著增加(P<0.05),青蒿组与卡托普利组均能明显减少房颤大鼠心肌β2-AR mRNA的表达(P<0.05),且两者之间差异无统计学意义。β2-AR蛋白的表达与mRNA的表达呈一致规律。结论β2-AR与房颤的发生关系密切,房颤大鼠β2-AR的表达明显增加;青蒿提取物能够有效地减少房颤大鼠心肌β2-AR的表达,这可能是青蒿抗心律失常的机制之一。Objective:To observe the expression and the Artemisia annua extract intervention on myocardial β2-adrenergic receptor mRNA through establishment of atrial fibrillation rats.Methods:40 rats were divided into the control group,the model group,the artemisia group and cartopil group.There were 10 rats in each group.The atrial fibrillation models were established with injection of Ach and CaCl2,intervened with artemisia.The cartopil was used in the control group.2 weeks before experiment,the artemisia group were fed with 1.25 g/kg,the cartopil group were fed with 3.25 mg/kg.The control and the model group were fed with NaCl.These indices were observed including the NF-κB mRNA expression and the cholestoral expresstion with PCR and western-blot.Results:Compared with the control group,model group rat cardiac β2-AR mRNA expression was significantly increased (P<0.05).The Artemisia annua group and captopril group could significantly reduce the atrial fibrillation rat myocardial β2-AR mRNA expression (P<0.05).There was no statistical difference between the two groups.β2-AR protein expression and mRNA expression were consistent with the law.Conclusion:β2-AR are closely related to the incidence of atrial fibrillation.Atrial fibrillation rat β2-AR expression is significantly increased.Artemisinic extract can effectively reduce fibrillation myocardial of β2-AR expression which may be one of artemisinic antiarrhythmic mechanism.
