摘  要:为了解不同性别刺五加中3-羟基-3-甲基戊二酰CoA还原酶(3-hydroxy-3-methylglutaryl coenzyme A reductase,HMGR)、甲羟戊酸焦磷酸脱羧酶(mevalonate diphosphate decarboxylase,MDD)和法尼基焦磷酸合酶(farnesyl diphosphate synthase,FPS)基因的表达特点及其对皂苷含量的影响,以GAPDH基因为内参照基因,通过实时定量PCR技术分析了不同性别类型刺五加中HMGR、MDD和FPS的表达量,并利用SPSS 17.0软件分析了其与刺五加总皂苷含量的相关性。结果表明,HMGR、MDD和FPS基因的最高表达量和皂苷含量的最大值均出现在短花丝类型刺五加中,中花丝类型的次之,长花丝类型的最低。HMGR、MDD和FPS的表达量和皂苷含量间存在显著正相关关系。HMGR、MDD和FPS是决定不同性别刺五加中皂苷含量高低的关键酶。To know the characteristics of expression of 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMGR), mevalonate diphosphate decarboxylase (MDD) and farnesy[ diphosphate synthase (FPS) genes in different gender types of Eleutherococcus senticosus and their influence on saponins content, taking GAPDH as a reference gene, quantitative real time PCR technique was used to analyze expression of HMGR, MDD and FPS in different sexual types of E. senticosus. The correlation between them and total saponins content was analyzed by using SPSS 17.0 software. The results indicate that the highest expression levels of HMGR, MDD and FPS are in the short filaments, followed by medium long filaments, and the lowest in the long filaments. Expression of HMGR, MDD and FPS has significantly positive correlation with saponins content. So HMGR, MDD and FPS are the key enzymes of determining saponins content in different gender types ofE. senticosus.
