摘 要:摘要:研究表明,严重的精子DNA损伤会导致男性不育、胚胎发育异常、高流产率以及降低辅助生殖技术的成功率。迄今为止,精子DNA的损伤与修复的机理尚不明了。本项目拟从精子形成和成熟的机制入手,以前期研究发现的具有体外优化精子作用的中草药麦冬为基础,进一步对麦冬皂苷及多糖进行体外精子培养及体内实验,分析比对精浆中活性氧含量与精子运动特征、形。态及顶体酶活性等的关系;鉴定中草药麦冬的体外活化精子有效成分。并通过大鼠应用麦冬皂苷和麦冬多糖建立动物实验模型,探讨麦冬体外活化精子和体内增强精子活性功能机制,了解精子DNA损伤及修复机理,为中药治疗少弱精症不育提供实验数据,提高男性不育患者的治疗效果。Recent years, attention has focused on the role of sperm nuclear DNA integrity in male factor infertility. Sperm DNA integrity is necessary for accurate transmission of genetic information and birth of healthy offspring. It is critical for normal embryogenesis, foetal well being and success of assisted reproductive techniques (ART). Some studies reported that severe DNA damage will lead male infertility, disordered embryonic development, high rates of miscarrige and increased risk of defects in the offspring. To date, the etiology of DNA damage is not very clear, and very little is known about the DNA repair mechanisms. In our previous study, we found that the extractives of ophiopogonis tuber have the effect of improving sperm motility in vitro. Our study is aimed to explore the effects of 0phiopogon saponins and ophiopogonpolysaccharides on sperm function and DNA in vitro and in vivo. Chemiluminescence, immunoblotting, long PCR, SCD, flow cytometry, and many other methods will be used to investigate the changes of acrosomat enzyme, , Caspase and DNA fragment in sperm. We are going to establish animal model to assess the influence of ophiopogon saponins and ophiopogonpolysaccharides on sperm function and DNA in vitro. This study may help to reveal the sperm DNA repair mechanisms and provide experimental data of a new kind of sperm medium.
