五味子含药血清中4种木脂素类成分的含量测定

摘　　要：目的:建立五味子含药血清中主要木脂素类成分含量测定的方法。方法:采用HPLC法测定,色谱柱为Lichrosphere C18(4.6 mm×250 mm,5μm),柱前加PhenomenexRDescription C18(4.0 mm×3.0 mm)保护柱,水-乙腈为流动相,梯度洗脱,流速为1.0 mL/min,柱温为30℃,检测波长为210 nm。结果:4种木脂素类成分五味子醇甲、醇乙、甲素和乙素分别在0.0512-0.7680μg、0.0540-0.8100μg、0.0123-0.1845μg和0.0398-0.5970μg范围内线性关系良好,相关系数r分别为0.9995、0.9996、0.9998和0.9996,含药血清中4种木脂素类成分的平均含量分别为19.3848、13.2624、1.3001和14.5842μg/mL。结论:该法准确、简便、专属性强,可用于五味子含药血清中主要木脂素类成分的测定。Objective： To establish a method for the assaying of main lignans in medicated serum of Schisandra chinensis. Methods： An HPLC method was set up, and Lichrosphere Cls column（4.6 mmμ50 mm, 5μm） was used and PhenomenexR De- scription Cis （g.0 mmx3.0 mm） was used as protective column. The gradient mobile phase was acetonitrile-water. The flow rate was 1.0 mL/min. The column temperature was 30 ~C and the detection wavelength was 210 nm. Results： The linear ranges of schisandrin, schisandrol B, deoxyschisandrin, and schisandrin B were respectively 0.0512-0.7680 Ixg, 0.0540-0.8100 ~g, 0.0123-0.1845 tLg, and 0.0398-0.5970 I.Lg, and the coefficient correlation （r） were 0.9995, 0.9996, 0.9998, and 0.9996, respec- tively. The average concentration of these 4 lignans in medicated serum were 19.3848, 13.2624, 1.3001, and 14.5842 ~g/mL, respectively. Conclusion： This method is accurate, easy, and specific for the assaying of lignans in medicated serum of Schisandra chinensis.
