摘  要:目的探索连续多次镉染毒后雄性小鼠的生殖毒性。方法将雄性NIH小鼠随机分为正常对照组、低、中、高剂量组共4组,每组6只动物,分别按0、0.17、0.34、0.68 mg Cd/kg的剂量腹腔注射氯化镉溶液,1次/d,连续5 d。染毒结束后观察120 d。雄鼠与雌鼠合笼测定交配能力,进行精子计数并观察精子畸形率;测定血清中睾酮、睾丸中镉含量;观察睾丸及附睾的病理变化。结果与正常对照组相比,中、高剂量组雄鼠附睾精子浓度和血清中睾酮水平均显著降低(P<0.05);低、中、高3个剂量组附睾中精子畸形率及睾丸中镉含量均显著升高(P<0.05);3个剂量组雄鼠的交配能力差异无统计学意义(P>0.05);部分动物的睾丸组织镜下发现边缘生精小管萎缩;各组动物的附睾中均未发现明显异常病理改变。结论雄性小鼠脱离接触镉后4个月仍可见生殖毒性。[ Objective] To explore the reproductive toxicity of male mice after cadmium exposure several times. [ Methods] Male NIH mice were divided into 4 groups : control group, low, medium, high dosage groups, with 6 animals in each group. Animals in these groups were exposed to cadmium chloride solution by intraperitoneal injection at the dosage of 0, 0. 17, 0. 34, 0.68 mg Cd/kg respectively for 5 d, once every day. After last injection, male animals were observed for 120 d. Then, every male mouse and a female one were put into a cage to measure the copulating capability of male animals. After that, the number of sperm was counted and the malformation was observed. Serum testosterone and testis cadmium were also measured. The pathological chan- ges in testes and epididymis were observed, too. [ Results] Comparing with male animals in control group, epididymis sperm concentration and serum testosterone of male mice in medium and high dosage groups were significantly lower ( P 〈 0.05 ), the epididymis sperm malformation rate and testis cadmium of male animals in the low, medium and high dosage groups were statistically higher ( P 〈 0.05 ). The copulating capability of all male mice in the 3 cadmium exposure groups was not statistically different ( P 〉 0.05 ). Atrophy was found in some seminiferous tubules at the edge of testis in some male mice. None apparent pathological changes were found in epididymis of all male mice. [ Conclusion] Although cadmium pretreated male mice were not exposed to cadmium for 4 months, their reproductive system may also be injured by cadmium at some extent.
