摘　　要：目的:研究HaCaT细胞经雷帕霉素处理后对不同剂量中波紫外线(UVB)照射的HaCaT细胞增殖活力的影响。方法:培养的细胞分为实验组(照射前雷帕霉素预孵育)和对照组(未加药组),两组各分5小组,均分别接受0、5、10、20和40 mJ/cm^2剂量的UVB照射。MDC染色观察自噬体表达水平。MTT法检测细胞接受UVB照射后12小时吸光度(OD)值。结果:雷帕霉素处理后细胞的自噬体表达明显高于对照组;在0、5、10和20 mJ/cm^2 UVB照射组的OD值均低于对照组,而在40 mJ/cm^2 UVB组OD值高于对照组,差异均有统计学意义(P<0.05)。结论:雷帕霉素预处理提高HaCaT细胞自噬水平后,对小剂量UVB照射后的HaCaT细胞增殖活力无保护作用,但对较大剂量UVB照射后HaCaT细胞增殖活力有保护作用。Objective： To determine the effects of UVB on the proliferation vigor of HaCaT cells after rapa mycin pretreatment. Methods： HaCaT cells were divided into experimental group （incubation with rapamycin before irradiation） and control group. Each group was divided into 5 sub-groups and treated with 0 mJ/cm2, 5 mJ/cm2, 10 mJ/cm2, 20 mJ/cm2 and 40 mJ/cm2 UVB irradiation. After 12 hours, the autophagy level after treating with rapamycin was evaluated by MDC staining method. The proliferation vigor of the cells was detec ted using MTF method. Results： MDC staining confirmed that rapamycin pretreatment significantly induced autophagy level. The cell proliferation ability （0, 5, 10 or 20 mJ/cm2 UVB） was significantly lower in experi mental groups than in control group. When the cells were irradiated with a dose of 40 mJ/cm2 UVB, however, the cell proliferation ability was increased compared with control group （P〈0.05）. Conclusion： After the au tophagy induced by pretreatment with rapamycine is increased, there is no protective role in the proliferation vigor of HaCaT cells at low dose of UVB irradiation, but, the protection is obviously increased in the prolifera tion vigor of HaCaT cells at a higher dose UVB irradiation.
