摘 要:目的观察补肾益气方对人早孕蜕膜基质细胞活性及其分泌白细胞介素10(IL-10)、干扰素γ(IFN-γ)及泌乳素(PRL)的影响。方法收集孕6~10周行人工流产的正常妊娠妇女蜕膜组织,体外分离、培养蜕膜基质细胞,分别以10%,20%补肾益气方含药血清处理细胞24 h、36 h、48 h,同时设立相应培养时间相同浓度的血清对照组,未添加血清组为空白对照组。MTT法检测补肾益气方对蜕膜基质细胞活性的影响。ELISA法检测细胞培养上清中IL-10、IFN-γ,化学发光法检测PRL分泌,流式细胞术检测IL-10R表达。结果 36 h起,20%中药组蜕膜基质细胞活性高于20%血清对照组(P<0.05),48 h时10%,20%中药组细胞活性均高于相应血清对照组(P均<0.05)。24 h起,10%,20%中药组分泌IL-10高于各对照组(P均<0.01);48 h时,20%中药组IL-10浓度高于10%中药组(P<0.05)并且中药组IL-10R表达高于各对照组(P<0.01),20%含药血清组IL-10R表达又高于10%含药血清组(P<0.05)。48 h之内,中药组IFN-γ水平与血清对照组相近(P>0.05)。24 h起,10%,20%中药组分泌的PRL高于各对照组(P均<0.01),并且20%中药组PRL水平高于10%中药组(P<0.01);48 h时2中药组之间PRL水平无显著性差异(P>0.05)。结论补肾益气方可能通过促进IL-10、PRL的分泌,上调IL-10受体表达,调控蜕膜基质细胞生长,达到保胎的目的。Objective It is to observe the influence of kidney-replenishing herb on the human first-trimester decidual stro- mal cells (DSC) secretion of interleukin - 10 ( IL - 10) , interferon - γ ( IFN - γ) and prolactin in early pregnancy. Meth- ods The decidual stromal cells were isolated from human villus by trypsin digestion and culture in vitro. Then the trophoblasts were treated with 10% and 20% kidney-replenishing herb for 24 h, 36 h and 48 h, respectively. Each exposure group was matched to same concentration serum control group which came from the same cells. The vitality of DSC was detected by MTT. The concentration of IL- 10, IFN -γ, prolactin in the culture medium was detected by ELISA and chemiluminescence reac- tion. The expression of IL - 10R was tested by flow cytometry assay. Results The vitality of DSC was significantly enhanced by 20% kidney-replenishing herb for 36 h compared with 20% serum control (P 〈 0.05) , and was stimulated role induced by 10% and 20% kidney-replenishing herb for 48 h compared with the same concentration serum control (P 〈 0.05). The secre- tion level of IL - 10 stimulated by 10% and 20% kidney-replenishing herb for 24 h to 48 h were increased significantly com- pared with those of serum control ( P 〈 0.01 ) , and IL - 10 stimulated by 20% kidney-replenishing herb for 48 h was increased significantly compared with those of 10% kidney-replenishing herb (P 〈0.05), and the expression of IL- 10R stimulated by kidney-replenishing herb was higher than that of control groups (P 〈 0.01 ) , meanwhile 20% kidney-replenishing herb signifi- cantly increased IL -10R level than those of 10% kidney-replenishing herb (P 〈 0. 05). The IFN -γ induced by kidney-re- plenishing herb is similar as the level of those serum control groups (P 〉 0.05).
