益气通络方对缺血性肾病早期大鼠肾组织及ICAM-1表达的影响

摘　　要：目的观察益气通络方对缺血性肾病早期大鼠的肾组织及其血管细胞间黏附分子-1(ICAM-l)表达的影响,探讨其防治缺血性肾病早期病变的机制。方法采用双侧肾动脉不全结扎术导致的双侧肾动脉狭窄建立缺血性肾病动物模型。SD雄性大鼠随机分为假手术组、模型组、预防组、药物组,于造模后14 d取材。采用HE染色及Masson染色观察肾组织病理学变化;应用免疫组化检测ICAM-1的表达。结果 HE染色及Masson染色结果显示模型组、预防组、药物组与假手术组比较都有不同程度的炎症及纤维化病灶,预防组与模型组比较,炎症及纤维化程度减轻;预防组、模型组与假手术组比较,ICAM-1表达增加;预防组与模型组比较,ICAM-1表达显著减少。结论益气通络方可能通过减少ICAM-1的表达,从而防治缺血性肾病的早期病变。Objective To observe the impacts of Yiqi Tongluo Formula on renal tissue and ICAM - 1 expression in the rats of early ischemic nephropathy and explore its mechanism on prevention and treatment of early ischemic nephropathy. Methods The incomplete ligation of bilateral renal arteries was adopted to induce bilateral renal arterial stenosis and establish the animal model of ischemic nephropathy. SD male rats were randomized into a sham - operation group, a model group, a prevention group and a medication group. The material was collected in 14 days after modeling. HE staining and Masson staining were used to observe the pathological changes in renal tissues. HTC was applied to detect the expression of ICAM - 1. Results In the observation with HE staining and Masson staining, compared with the sham -operation group, the inflam- matory disorders and fibrosis foci were displayed to different extents in the model group, the prevention group and the medication group. Compared with the model group, the degree of inflammatory disorders and fibrosis were alleviated in the prevention group. Compared with the sham - operation group, ICAM - 1 expression was increased in the prevention group and the model group. Compared with the model group, ICAM - 1 expression was reduced significantly in the prevention group. Conclusion Yiqi Tongluo Formula prevents and treats early ischemic nephropathy probably via reducing the expression of ICAM - 1.
