摘 要:为了研究天然化合物丁香酚对番茄侧根生长的影响,本试验以番茄幼苗为材料,研究不同浓度丁香酚和硫化氢(H2S)供体硫氢化钠(NaHS)对番茄幼苗侧根生长的影响,以及H2S清除剂亚牛磺酸(HT)对丁香酚和NaHS调控侧根发生过程的影响。同时,根据番茄根细胞内Ca2+的响应情况,研究丁香酚调控侧根发生可能的信号途径。结果显示:(1)丁香酚处理能显著促进番茄幼苗侧根的生长,并呈现出浓度和时间效应,其中30μmol/L丁香酚促进侧根生长效果最为显著;H2S供体NaHS可以增加番茄侧根数目,15μmol/L NaHS是促进番茄侧根生长的最佳浓度。(2)H2S清除剂HT可以抑制NaHS和丁香酚促进侧根生长效应,表明丁香酚促进侧根生长效应与细胞内H2S信号途径有关。(3)NaHS和丁香酚处理都可显著提高番茄幼苗根细胞内Ca2+含量,而加入HT后则抑制了丁香酚诱导Ca2+积累的效应。上述结果表明丁香酚促进侧根生长过程是通过调控H2S信号,进而导致根细胞内Ca2+的累积,最终促进侧根的生长。To determine the effect of eugenol, a naturally plant-derived compound, on the growth of tomato lateral root, different concentrations of eugenol and Naris (a donor of hydrogen sulfide H2S) were applied to lateral root of tomato seedlings. The effect of HT ( a H2S scavenger) on the eugenol- and NariS-regulated growth of lateral root were identified as well. According to the response of the tomato root intracellular Ca2 + , the possible signaling pathways regulating the lateral root by eugenol was studied. The main results were as follows. (1) Eugenol promoted the growth of tomato lateral root in both concentration- and time-dependent manners, and 30μmol/L eugenol showed the greatest effect. Naris could also stimulate the growth of lateral root in both concentration- and time-dependent manners, and 15 μmol/L Naris showed the greatest effect. 2) Removing H2 S by the addition of HT in treatment solution could inhibit the promoting effect of eugenol and Naris on lateral root, suggesting that the promoted effects of eugenol was associated with H2S in the cell signaling path-ways. (3) Eugenol and NariS stimulated the accumulation of Ca2+ in tomato root. However, HT blocked the accumulation. These results suggested that engenol promo- ted the growth of tomato lateral root by regulating H2S sig- nal followed by cytosolic Ca2+ accumulation in root cells.
