摘 要:以山葡萄转色期果皮为供试材料,从构建的山葡萄转色期果皮cDNA文库中选取与花色苷生物合成相关的调节基因进行cDNA全长克隆和生物信息学分析。结果表明,转录因子MYBA3基因的cDNA全长序列为764 bp,编码250个氨基酸。与其他物种相关基因的氨基酸序列的多重比较结果表明,它与欧亚种葡萄(Vitis vinifera L.)相似性达到97%。由该基因推导的蛋白质不具有信号肽结构,具有一定的亲水性,二级结构以α-螺旋和无规则卷曲为蛋白质最大量的结构元件。这为阐明该基因在山葡萄花色苷生物合成及调控中的作用提供了依据。Based on cDNA library of Vitis amurensis pericarp at veraison, the full-length cloning and bioinformatics analysis of regulator gene concerning anthocyanins synthesis was conducted. The full-length sequence of transcription factor MYBA3 obtained from Vitis amurensis pericarps at veraison was 764 bp, encoding 250 amino acids. Allignment of amino acid sequences with relevant genes from other species showed that it shared, 97% similarity with V. vinifera L.. The de- duced protein was hydrophobic and signal peptide free, and its secondary structure was dominated by a-helix and random coil. These results would provide an basis for identifying the role of MYB gene in biosynthesis and regulation of anthocya- nins in F. amurensis.
