摘 要:研究制备以聚乙烯吡咯烷酮为载体的阿霉素纳米粒,提出用HPLC-荧光色谱法测定聚乙烯吡咯烷酮-阿霉素纳米粒体系中阿霉素含量,并采用该方法测定了纳米粒的包封率和载药量.结果表明,制备得到的阿霉素纳米粒粒径小,球形度好;HPLC-荧光色谱法对聚乙烯吡咯烷酮-纳米粒体系中的阿霉素含量测定具有专属性强、方便快捷的特点;阿霉素在0.08~8 μg/mL范围内线性关系良好(R2 =0.9999),回收率高;阿霉素纳米粒载药量为(2.92±0.09)%,包封率为(83.21±2.17)%.Objective: Doxorubicin- loaded nanoparticles, using doxorubicin (ADM) as model drug, polyvi-nylpyrrolidone (PVP) as matrix material, were prepared in this paper. Methods: More over, the drug loading and encapsulation efficiency were measured by high performance liquid chromatography with fluorescence detection (HPLC -FLD) . Results: The results showed that the nanoparticles exhibited a spherical shape and a small size from the observation of SEM. Calibration curve of doxorubicin in the system was constructed in the concentration range 0. 08 - 8.0 μg/mL ( R2 = 0. 9999 ) . Their drug - loaded amount and encapsulation efficiency were (2. 92 ±0. 09) % and (83.21 ± 2. 17) % respectively. Conclusions: The consequence indicated that the HPLC -FLD was fine to detect the concentration of doxorubicin in nanonarticles.
