化疗药物环磷酰胺对家蚕血球密度和精细胞发育的影响

摘　　要：为探讨家蚕作为化疗药物和免疫抑制药物试验的模型动物的可能性,研究不同剂量环磷酰胺对家蚕血球密度、5种血球比率、精母细胞减数分裂及精细胞变形过程的影响。结果表明,环磷酰胺能显著降低家蚕血球密度,抑制血球增殖;对原白血球和颗粒细胞的比率基本无影响,但使浆细胞比率极显著升高,使小球细胞和拟绛色细胞的比率下降;对精母细胞减数分裂和精子变形成熟过程也具有极为显著的抑制作用。初步确定环磷酰胺抑制家蚕血球增殖和精细胞发育的适宜注射剂量为每克蚕体质量200μg(即200μg/g),适宜添食剂量为500μg/g,高于对哺乳动物的有效剂量。研究结果证明环磷酰胺对低等动物家蚕的细胞分裂和增殖的抑制作用与在高等动物中的作用相同或相似,因此,家蚕有望成为环磷酰胺类似化疗药物、免疫抑制药物和不育模型药物的实验动物。To inquire into the possibility of using silkworm as a model animal for testing chemotherapeutic drugs and im- munosuppressive drugs, the effects of different doses of cyclophosphamide on the density of silkworm blood cells, the proportions of 5 blood corpuscles, spermatocyte meiosis and sperm cell formation were investigated. The results show that cyclophosphamide can significantly reduce the density of silkworm blood cells and inhibit proliferation of blood cor- puscles. Cyclophosphamide has no effect on proportions of prohemocytes and granulocytes, but extremely significantly increases the proportion of plasmatocytes. It reduces proportions of spherulocytes and oenocytoids, and also has ex- tremely significant inhibitory effect on spermatocyte meiosis, sperm formation and maturation. It is preliminarily deter- mined that, to inhibit silkworm blood cell proliferation and sperm cell development, the appropriate dose of cyclophos- phamide via injection was 200 pg per gram silkworm body （i. e. 200 IJg/g） and that via oral administration was 500 ~g/g, higher than the effective dose to mammals. The above results showed that cyclophosphamide has the same or similar role in inhibiting cell division and proliferation between lower animal silkworm and higher animals, so silkworm has the potential to become the experimental animal model for testing cyclophosphamide-like chemotherapeutic drugs, immu- nosuppressive drugs and drugs to treat sterility.
