摘 要:目的观察益肾通络方对链脲佐菌素(STZ)诱导糖尿病模型大鼠内皮素(ET)、一氧化氮(NO)、血栓烷素B2(TXB2)、6-酮-前列腺素1α(6-keto-PGF1α)水平的影响。方法用STZ诱导糖尿病模型大鼠,随机分为模型组、葛根素组以及益肾通络方大剂量组、益肾通络方小剂量组,并设正常对照组。各组干预6周后,检测各组对大鼠体质量、血糖以及血浆中ET、NO、TXB2、6-keto-PGF1α含量。结果与正常对照组比较,模型组大鼠体质量明显减轻,血糖升高,血浆中ET、TXB2含量明显升高,NO、6-Keto-PGF1α含量明显减低,差异有统计学意义(P<0.05,P<0.01)。与模型组比较,益肾通络大剂量组体质量增加,NO、6-Keto-PGF1α含量升高、ET、TXB2含量降低,差异有统计学意义(P<0.05,P<0.01);益肾通络小剂量组ET含量降低、6-Keto-PGF1α含量升高(P<0.01);葛根素组空腹血糖降低,ET、TXB2含量降低,6-Keto-PGF1α含量升高,差异有统计学意义(P<0.05,P<0.01)。与葛根素组比较,益肾通络大剂量组ET、TXB2含量下降,差异有统计学意义(P<0.05)。结论益肾通络方能够改善糖尿病模型大鼠NO、ET、TXB2、6-Keto-PGF1α的含量,从而保护血管内皮功能。Objective To observe the effect of "Yishen Tongluo Formula"on the levels of ET, NO, TXB2 and 6-keto-PGFt~ in diabetes induced by STZ in rats. Methods The rats were randomly divided into model group, puerarin group, low dose of " Yishen Tongluo Formula" group, large dose of " Yishen Tongluo Formula" group and normal control group. After 6 weeks intervention, the body weight, blood glucose and the contents of NO, ET, TXB2 , 6-Keto-PGF1α in blood plasma were detected. Results Compared with normal control group, the weight of rats in model group was obviously reduced, the content of ET and TXB2 were increased, and the content of NO and 6-Keto-PGF1α were decreased(P 〈 0.05, P 〈 0.01 ). Compared with model group, weight of rats, the contents of NO and 6-Keto-PGF1α, in large dose group was increased, and the contents of ET and TXB2 in high dose group were decreased markedly( P 〈 0.05, P 〈 0. O1 ) ; the ET content was decreased, 6-Keto-PGF1α content was increased(P 〈 0.01 ) ; the FBG and the contents of ET and TXB2 were decreased, but the 6-Keto-PGF1α content was increased in puerarin group, with significant difference between puerarin group and model group(P 〈0.05, P 〈0.01 ). Compared with puerarin group, the contents of ET and TXB2 in large dose group were reduced significantly ( P 〈 0.05 ). Conclusion "Yishen Tongluo Formula" can protect the vascular endothelial function by improving the contents of NO, ET, TXB2 and 6-Keto-PGF1α.
