摘  要:目的探讨芪丹通脉片对慢性脑缺血大鼠海马MDA、SOD表达的影响及其可能机制。方法采用双侧颈动脉结扎方法制作慢性脑缺血模型。将健康雄性SD大鼠随机分为空白对照组、模型组、假手术组、芪丹通脉片低剂量组、芪丹通脉片中剂量组、芪丹通脉片高剂量组、阳性对照尼莫地平组,HE染色观察海马CA1区神经元形态学改变,并检测海马MDA、SOD的表达。结果与对照组比较,模型组大鼠可见海马神经元呈现出典型的神经病理性改变,海马CA1区神经细胞数量减少、固缩。芪丹通脉片可降低海马MDA含量,提高SOD活性,与模型组比较有显著性差异(P<0.05),且呈剂量依赖性。结论氧化应激可能参与慢性脑缺血所致的海马神经元丢失,推测芪丹通脉片可通过抗氧化应激机制发挥一定的神经保护作用。Objective To observe the effects of QiDanTongMai Tablets (TDTMT) on MDA and SOD expression in the hippocam- pus of rats with chronic cerebral ischemia and to explore its mechanisms. Methods The chronic cerebral ischemia rat's model was made with permanent ligation of the bilateral carotid arteries. Male - SD rats were divided into seven groups as follows : normal group, control group, model group, sham - operation, low dosage TDTMT, moderate dosage TDTMT, high dosage TDTMT, Nimodip- ine group. HE staining method was used to observe the morphological changes of hippocampal CA1 area. MDA and SOD expression in the hippocampus of rats was detected. Results Compared with normal group, hippocampal neurons in rats of model group showed typical neuropathologic lesion, especially neurons decreasing and pyknosis in hippocampal CA1 area. Compared with the model group, TDTMT could reduce the level of MDA and increase SOD activity, which was a significant difference ( P 〈 0.05 ) and was related to TDTMT dosage. Conclusion Oxidative stress may be involved in hippocampal neuronal loss induced by chronic cer- ebral ischemia. TDTMT may play a neuroprotective effect through anti - oxidative stress mechanism.
