摘  要:目的通过比较水飞蓟宾经乳糖修饰前后的护肝效果,评价水飞蓟宾乳糖修饰对小鼠急性酒精肝损伤保护作用的影响。方法采用预防给药方式,灌胃给予50%乙醇建立急性酒精肝损伤模型,分别测定水飞蓟宾(Sil)、水飞蓟宾乙酰乳糖苷(Sil-LacAcO8)和水飞蓟宾乳糖苷(Sil-Lac)组小鼠血清天冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)活性和甘油三酯(TG)、胆固醇(TC)含量以及小鼠肝匀浆超氧化歧化酶(SOD)活性和甘油三酯(TG)、丙二醛(MDA)、还原性谷胱甘肽(GSH)水平,油红O染色观察肝脏病理形态学变化。结果与模型组比较,水飞蓟宾、水飞蓟宾乙酰乳糖苷和水飞蓟宾乳糖苷组的肝脏病理损伤减轻,SOD活性显著升高(P<0.01),GSH水平明显升高(P<0.05,P<0.01,P<0.01)。水飞蓟宾乙酰乳糖苷和水飞蓟宾乳糖苷组血清AST和ALT活性明显降低(P<0.01、p<0.05),肝匀浆MDA水平降低(P<0.01)。与水飞蓟宾组比较,水飞蓟宾乳糖苷组血清AST和ALT活性明显降低(P<0.01),血清TC明显降低(P<0.05);水飞蓟宾乙酰乳糖苷组血清AST活性明显降低(P<0.05),SOD活性明显升高(P<0.05)。结论水飞蓟宾经乳糖修饰后对小鼠急性乙醇所致肝损伤具有更好的保护作用。Objective Evaluation of the effect of Silybin -23 -O- lactoside (Sil- Lac), acetylated lactoside( Sil -LacAcO8 ) on acute alcohol- induced liver injury in mice by comparing the hepatoprotective activity between silybin and silybin lactoside. Methods The method was as follows: 50% ethanol was administered orally to establish acute alcoholic liver injury model after 15 days continuous intraperitoneal drug injection. Hepatotoxicity was evaluated by measuring the serum levels of ferase ( AST), alanine aminotransferase ( ALT), cholesterol ( TC ), triglyceride ( TG ) and hepatic triglyceride ( TG), malondialde- hyde (MDA) as well as the tissue levels of antioxidant enzyme such as superoxide dismutase (SOD) and reductase glutathione (GSH) in mice, and histopathological change was assessed by hematoxylin and eosin staining. Results The treatment with Sily- bin, Sil - LacAcO8 and Sil - Lac could attenuation the changes of histopathological, increase the activities of SOD ( P 〈 0.01 ), and GSH( p 〈 0.05, P 〈 0.01, P 〈 0.01 ) in liver tissues compared with alcohol - induced model control(P 〈 0.01 ), and the treatment with Sil - LacAcO8 and Sil - Lac could decrease serum AST, ALT ( P 〈 0.01, P 〈 0.05 ) and MDA ( P 〈 0.01 ) levels significantly in plasma compared with alcohol - induced model control. The treatment with Sil - Lac could decrease serum AST, ALT activities ( P 〈 0.01 ) and TC level ( P 〈 0.05 ) ; The treatment with Sil - LacAcO8 could decrease serum AST activity ( P 〈 0.05 ) and increase the activity of SOD (P 〈 0.05 ) compared with Sil administration. Conclusion The protective effect of Sil - Lac, Sil - LacAcO8 on acute alcohol - induced hepatotoxicity was better than Silybin.
