摘 要:为了研究铜绿微囊藻生长过程中毒素的产生与释放特性,在实验室固定条件下培养铜绿微囊藻,以细胞计数反映其生物量,用沸水法提取细胞内毒素,分别测定胞外、胞内微囊藻毒素LR的质量浓度。结果表明,在铜绿微囊藻的不同生长阶段,细胞外与细胞内毒素有不同的变化趋势。在其整个生长阶段,细胞内微囊藻毒素LR与生物量呈较好的正相关关系;细胞外MC-LR质量浓度随培养时间的增加不断上升,细胞数达到最大后,MC-LR质量浓度增加速率变大。The present paper is aimed at studying the production and release characteristic features of the toxic microcystin-LR of Microcys- tis aeruginosa in the growing stage. For our research purpose, we have cultivated a group of toxigenic Microcystis aerugirtosa under the certain laboratory conditions. In proceeding with our research, we have carefully assessed the biomass of the Microcystis aeruginosa by counting the number of the cells with the automatic cell counter: the CountStar, and centrifuged the cell suspension for the supernantant and algae cells, respectively. And, next, we have managed to enrich the supernantant by using the CIs solid phase extraetion(SPE) car- tridges for finding the extracellular microcystin-LR. And, next, we resuspended the cells with deionized water and tried to extract the toxin microcystin-LR from the cells with the boiling water. Actually, each sample has to be extracted in the boiling water for 20 minutes. Furthermore, it is necessary to centrifuge the sample and enrich the supernatant by using the CIs SPE cartridges to detect the intracellular microcystin-LR. And, afterwards, the cartridges had to be rinsed in 10 mL deionized water with 20% methanol, and then elute MC- LR by using 90% methanol with 0.1% TFA. The eluents could then be dried under nitrogen at 40 ℃. And, consequently, the rest sub- stances could be dissolved in 0.5. mL methanol (two times ) and transferred into autosampler vial. When dried again in the stream ofnitrogen, diluted with 50% methanol to 200 μL, they could be found by high-performance liquid chromatographic (HPLC). The results of our experiments prove that during the different growing stages of Mi- crocystis aeruginosa, the changing trends of the extracellular & intra- cellular microcystin-LR content can be various. It can also be found that the intracellular microcystin-LR content took on a perfect linear relationship with the biomass of the Microcystis aeruginosa in the whole growing stage, with their related coefficient being 0.915 2
