摘  要:目的观察针刺对去卵巢骨质疏松症模型大鼠雌二醇、生长素和骨密度的影响。方法选取60只3月龄SD健康雌性大鼠,按随机数字表法分为手术组和假手术组,手术组40只,假手术组20只。采用双侧卵巢切除法复制成质疏松大鼠模型,造模3个月后经双能X线法检测BMD,手术组大鼠腰椎和股骨的骨密度较假手术组明显降低(P〈0.01),表明骨质疏松模型造模成功。造模成功后,从假手术组选取10只大鼠设为对照组,从手术组选取30只大鼠按随机数字表法分为模型组、针刺组和雌激素组,每组10只。其中,针刺组给予电针刺激,雌激素组皮下注射苯甲酸雌二醇,对照组和模型组不作任何处理。连续干预3个月后行骨密度(BMD)检测,然后处死大鼠,摘除眼球取血,检测血清雌二醇、生长素等指标。结果血清生长素:针刺组[(399±163)pg/ml]和雌激素组[(276±92)pg/ml]与模型组[(546±98)pg/ml]比较,均有明显降低(P〈0.01);血清雌二醇:针刺组[(128.02±62.28)pg/ml]和雌激素组[(182.89±42.01)pg/ml]与模型组[(72.10±19.83)pg/ml]比较,均有明显提高(P〈0.05);腰椎骨密度:针刺组[(0.212±0.019)g/cm^2]和雌激素组[(0.231±0.021)g/cm^2]与模型组[(0.191±0.012)g/cm^2]比较,均有明显提高(P〈0.05),但针刺组对生长素和腰椎骨密度的作用不及雌激素组(P〈0.05)。结论针刺疗法能明显提高去卵巢骨质疏松症模型大鼠腰椎骨密度和血清雌二醇的水平,下调血清生长素的水平,但整体作用不及雌激素。Objective To observe the effect of acupuncture on ghrelin, estradiol (E2) and bone mineral density (BMD) in postmenopausal osteoporosis rats. Methods According to the random number table method, 60 female Sprague-Dawley rats aged 3 months, were randomly divided into surgery group (40 rats) and sham group (20 rats). The osteoporosis model was established by bilateral ovarieetomy. Three months after the operation, a signifi- cant decrease in BMD of lumbar vertebrae and femur of the model rats as measured by dual energy X-ray absorption- metry against the sham group indicated successful establishment of the osteoporosis model. Then surgery group was randomly divided into a model subgroup, a acupuncture subgroup and an estrogen subgroup, with 10 rats in each sub- group. The acupuncture subgroup was treated with electroacupuncture, estrogen subgroup with subcutaneous injec- tion of estradiol benzoate, sham group and model subgroup without any treatment. After 3 months of intervention all the rats were sacrificed, BMD of the lumbar vertebrae and femur was measured, ghrelin and E2 were determined. Results Serum ghrelin in acupuncture subgroup (399 pg/ml) and estrogen subgroup (276 pg/ml) were signifi- cantly lower ( P 〈 0. 01 ) than that in model subgroup (546 pg/ml) ; serum E2 in acupuncture subgroup ( 128.02 pg/ml) and estrogen subgroup ( 182.89 pg/ml) increased significantly ( P 〈 0.05 ) than that in model subgroup(72.10 pg/ml) ;lumbar vertebrae BMD in acupuncture subgroup (0. 212 g/cm2 ) and estrogen subgroup (0. 231 g/em2 ) increased significantly ( P 〈 0.05 ) than that in model subgroup ( 0. 191 g/cm2 ) , but in acupunc- ture subgroup the effect on improving the level of E2, ghrelin and lumbar vertebrae BMD was less than that in estro- gen subgroup (P 〈 0.05). Conclusions Acupuncture intervention can improve lumbar vertebrae BMD and ser- um E2 level of postmenopausal osteoporosis rat model, down regulate serum ghrelin level, but the overal
