摘 要:目的探讨Foxn1在小鼠胸腺增龄性萎缩中的作用。方法 SPF级C57BL/6小鼠分成老龄组(18个月)、中龄组(12个月)、中青龄组(6个月,4个月)、青龄组(1个月),每组雌雄各6只。无菌取胸腺,比较小鼠胸腺重量、胸腺指数及胸腺细胞数;应用Real-time PCR技术检测胸腺组织中Foxn1基因的表达。结果小鼠胸腺重量及胸腺细胞数随年龄增长显著减少(P<0.05,P<0.01);C57BL/6小鼠胸腺Foxn1相对表达量分别为:1月龄(0.909±0.119),6月龄(0.689±0.108),12月龄(0.240±0.063),18月龄(0.047±0.010),表现为随年龄增长显著下调(P<0.05)。结论Foxn1参与了胸腺的发育、成熟和退化萎缩各个阶段,Foxn1表达降低破坏了胸腺微环境,导致胸腺增龄性萎缩。Objective To investigate the role of Foxn1 on the age-related thymic involution in mouse.Methods SPF grade C57BL/6 mice were divided into older age (18 months),middle-aged (12 months),younger age (6 months and 4 months),(1 months)groups,male and female 6 mice of each group.The thymus weight,thymus index and the number of thymocytes were compared and Foxn1 gene expression in thymus tissue was tested by Real-time PCR.Results The mouse thymus weight and thymocyte age were decreased along with the age growth.C57BL / 6 mice the thymus The Foxn1 relative expressions were 0.909 ± 0.119 (young age group),0.689 ± 0.108 (younger age group),0.240 ± 0.063(middle-aged group),0.047 ± 0.010(older age group),and there were significant differences among four groups.Conclusions Foxn1 is involved in all stages of the development,maturation and involution of the thymus,the lower expression of Foxn1 reduce the destruction of the thymic microenvironment,which lead to the age-related thymic involution.
