摘  要:目的:观察环磷酰胺和地塞米松对小鼠肠道黏膜免疫的抑制作用。方法:筛选地塞米松和环磷酰胺的给药天数和剂量;CCK-8检测肠系膜淋巴结(Mesenteric lymph nodes,MLN)细胞转化增殖;流式细胞术检测MLN及派氏结(Peyer'sPatch,PPs)中T、B淋巴细胞的数量及活化比例;ELISA试剂盒检测肠道sIgA的分泌。结果:地塞米松40 mg/kg和环磷酰胺100 mg/kg腹腔注射给药一次,有效降低小鼠胸腺、脾脏脏器指数和减少派氏结个数,抑制MLN细胞增殖;与正常组相比,地塞米松降低MLN和PPs中T细胞比例,抑制T、B细胞的活化;而环磷酰胺降低MLN和PPs中B细胞的比例,对MLN中B细胞活化有明显的抑制作用;地塞米松和环磷酰胺均降低肠道sIgA的分泌。结论:地塞米松和环磷酰胺皆能对肠道黏膜免疫产生抑制作用,但地塞米松对MLN和PPs中T细胞抑制作用较强,而环磷酰胺对B细胞的抑制作用较强。Objective: To study the immunosuppressive functions induced by Cyclophosphamide (CY) and Dexamethasone (DEX) in intestinal mucosa in vivo in mice. Methods:Firstly screening out the administration dosages and days of DEX and CY; Cell proliferation was detected by CCK-8 in mesenterie lymph nodes (MLN) ; the activation and proportion of T and B lymphocytes were de- tected by Flow Cytometry in MLN and Peyer's patch (PPs) ; intestinal slgA was detected by ELISA. Results:DEX (40 mg/kg) and CY ( 100 mg/kg) intraperitoneal injection once could effectively reduce the thymus and spleen Organ Index, the number of PPs, and inhibit lymphocyte proliferation in MLN ; compared with the normal group, DEX reduces the proportion of T cells and inhibits the acti- vation of both T and B cells in MLN and PPs ; CY could reduce the proportion of B cells in the MLN and PPs, and significantly inhibit the activation of B cell in MLN; DEX and CY decreased secretion of intestinal slgA. Conclusion:Both DEX and CY could inhibit in- testinal mueosal immunity significantly, however DEX has strongly inhibited T cells and CY inhibited B cells.
