摘 要:目的:研究五味子藤茎木脂素和多糖对急性酒精肝损伤的保护作用。方法:除空白对照组外,其余各组小鼠酒精灌胃给药造成急性肝损伤。应用酶标法测定小鼠血清中谷氨酸转氨酶(ALT)、天门冬氨酸转氨酶(AST)活性和甘油三酯(TG)浓度,并测定小鼠肝脏中超氧化物歧化酶(SOD)活性及丙二醛(MDA)浓度。结果:与模型组相比,五味子木脂素和多糖高、中、低剂量能不同程度地降低血清中ALT、AST活性及TG浓度,升高肝脏中SOD的活性并抑制MDA浓度的上升。其中,高剂量的作用效果与联苯双酯作用相当,但二者对各指标的作用程度不同。结论:五味子藤茎木脂素和多糖对酒精诱导的急性肝损伤有一定的保护作用,二者的作用机制可能与抗氧化作用有关。Objective : To observe the protective effects of lignans and ploysaccharides from caulis of Schisandra chinensis against acute injmy induced by ethanol in mice. Methods: Except the blank control group, the other groups were made the liver injury model by administering ethanol. The hepatoprotective effects were evaluated by assessing alanine aminotransferase (ALT), aspartate aminotransferase (AST) levels and triglyceride (TG) content in serum, superoxide dismutase (SOD) activity and content of malondialdehyde (MDA) in liver using enzyme label method. Results: Compared with the model groups, the three different dose groups of lignans and polysaecharides reduced ALT, AST levels and TG content in serum, simultaneously, increased the activity of SOD and reduced the MDA content in liver to varying degrees. Among them, the hepatoprotective effect of high dose group was equal to that of Bifendatatum (Positive drug group). Conclusion: Lignans and ploysaccharides from caulis of Schisandra chinensis posses the protective effect on ethanol-induced acute hepatic injury. The action mechanism may be related to their antioxidant capabilities.
