摘  要:目的:观察冠心康颗粒对高同型半胱氨酸血症致兔动脉粥样硬化的作用及对血管炎性因子VCAM-1、MCP-1表达的影响。方法:24只雄性新西兰大耳兔随机分成3组,空白对照组、同型半胱氨酸模型组、冠心康治疗组,饮食量为100 g/d。分别于第1、6、10周兔耳中央动脉取血检测血浆中同型半胱氨酸的含量。第10周末处死动物,动脉组织HE染色观察主动脉病理形态学变化,免疫组织化学技术检测动脉血管中NF-κB的表达,RT-PCR方法检测动脉组织中VCAM-1、MCP-1的基因表达。结果:第6、10周末模型组血浆中同型半胱氨酸浓度显著高于同期空白对照组(P<0.05)。第10周末模型组光镜下可见血管内膜早期动脉粥样硬化样改变;主动脉内皮细胞NF-κB p65、VCAM-1以及MCP-1表达增强;动脉组织中VCAM-1、MCP-1的mRNA表达高于空白对照组(P<0.05)。治疗组与模型组相比血浆中同型半胱氨酸浓度显著降低(P<0.05),主动脉内膜损伤程度减轻;NF-κB p65、VCAM-1以及MCP-1蛋白表达减少,动脉组织中VCAM-1、MCP-1的mRNA表达减少。结论:冠心康颗粒具有抗动脉粥样硬化的作用,该作用与降低血浆中同型半胱氨酸的浓度,抑制主动脉内皮细胞NF-κB活性,从而抑制炎症因子VCAM-1和MCP-1表达有关。Objective : To explore the effects of Guanxinkang granules on atherosclerotic lesion induced by hyperhomocysteinemia in rabbits and its effects on vascular cell adhesion molecule-1(VCAM-1) and monoeyte chemoattractant protein-1(MCP-1) gene expression in aorta. Methods: Twenty-four male rabbits were randomly divided into three groups, the control group, the homocysteine (Hcy) model group and the treatment group. Blood samples of rabbits were collected after 1, 6 and 10 weeks of experimental diets for measurement of plasma Hcy. The aorta was removed at the end of the protocol for investigation of nuclear factor- κB 65 protein, VCAM-1 and MCP-1 expressions were tested by immunohistochemistry, VCAM-1 and MCP-1 gene expression by RT-PCR and sections were made for light microscopy to observe the vascular pathomorphologic changes 10 weeks after experiment. Results : The level of plasma Hcy in the model group was significantly higher (P〈0.05)at 6 and 10 weeks after experiment than that in the control group. Simultaneously, early stage of atherosclerosis in tunica intima could be easily observed and NF- κB 65, VCAM-1 and MCP-1 of aorta in model group expressed markedly higher than those in the control group at the end of 10 weeks. While the level of plasma Hcy was decreased and aortic intimal changes in pathomorphology were attenuated in treatment group compared with those in model group. And at the same time, the protein expression of NF- κB p65, VCAM-1 and MCP-1 and gene expression of VCAM-1 and MCP-1 were also decreased in the treatment group ( P〈0.05 ). Conclusions : The results suggest that Guanxinkang granules has anti-atherosclerosis effect by reducing plasma Hcy level and inhibiting NF- κB, VCAM-1 and MCP-1 expressions.
