干式血红蛋白检测系统的研制与初步评估

摘　　要：目的 研制干式血红蛋白检测系统并进行性能评估,从而实现应急条件下采供血的快速检测.方法 利用红细胞破碎试验确定溶血剂及其溶度,通过全谱扫描确定血红蛋白分析仪所需波长,结合氰化高铁血红蛋白法测定结果绘制血红蛋白分析仪的标准工作曲线.选择不同浓度梯度Triton-100和碳酸钠包被液制作成试条,检测10份全血的血红蛋白含量,选择最佳包被液配方.对所研制的干式血红蛋白检测系统进行线性、重复性、准确性和稳定性测试.结果 干式血红蛋白检测系统的检测波长为540 nm,以浓度为20 g/L Triton-100和21.2 g/L碳酸钠的包被液制作试条.本系统的检测结果与迈瑞BC-5800检测结果相关性好,准确度在±5％以内,重复性CV值为3.58％,血红蛋白浓度在40～240 g/L范围内线性良好,试条在22个月内检测结果有效.结论 干式血红蛋白检测系统能满足临床及应急条件下血红蛋白检测的要求.Objective To prepare and evaluate the detection system for dry-type hemoglobin so as to to defect blood collection quickly in emergency conditions. Methods Hemolytic agents of appropriate concentration were measured, using the red blood cell crushing test. Standard work curves of the hemoglobin analyser were drawn with the references from the wavelengths of hemoglobin analyzer detected by the qualitative spectral scanning and the detection results of cyanmethemoglo- bin. Different concentration gradients of Triton-100 and solution of sodium carbonate peridium were chosen to make reagent strips, which were used to detect the hemoglobin content in 10 samples of whole blood so as to choose the optimal prescription of peridium solution. The linearity, repeatability, accuracy and stability of dry-type hemoglobin detection system were tested. Results The dry-type hemoglobin detection system included detective wavelength of 540 nm and reagent strips by using 20 g/L Triton-100 and concentration of 21.2 g/L sodium carbonate peridium. The result of the system was in good connection with BC-5800. The accuracy was within ＋5%. The CV value of repeatability was 3.58%. The better linear range of hemo- globin concentration was within the range of 40 - 240 g/L. The expiration date of reagent strip was within 22 months. Con- clusion The system can be used in all kinds tests of hemoglobin for clinical purpose and emergency cases.
