摘 要:克雷伯氏菌是1,3-丙二醇(1,3-PD)生产菌。以产酸克雷伯氏菌2-1为出发菌株,经硫酸二乙酯(DES)诱变处理,运用质子自杀法选育,从含0.2 mol/L NaBr-NaBrO3的初筛平板上选出48株单菌落,然后结合培养基优化后的摇瓶发酵复筛,获得6个产酸突变株,其中Y-37具有较高的1,3-丙二醇转化率。经过初筛、复筛和传代实验,表明其是稳定的突变株。对菌株Y-37在自动发酵罐上进行批式发酵,结果显示:突变株产酸量大幅降低,而1,3-丙二醇的产量则显著增加。Y-37的1,3-丙二醇产量由出发菌株的10.67g/L升至18.23g/L。Klebsiella pneumoniae is 1,3-propanediol (1,3-PD) production of bacteria. After primary screening, secondary screening and passage experiments show that it is the stability of the mutant strains. Strain Y-37 in the automatic batch fermentation on the fermentation. The results show: mutants significantly reduce acid production, and 1, 3 - propanediol production increased significantly. Y-37 for 1,3-propanediol production by a strain of 10.67g/L increased to 18.23g/L.
