宫颈鳞癌组织中pSTAT3、Cyclin D1、VEGF表达变化及意义

摘　　要：目的观察宫颈鳞癌组织中磷酸化信号转导和转录激活因子3(pSTAT3)、细胞周期素D1(Cyclin D1)及血管内皮生长因子(VEGF)蛋白的表达变化,并探讨其临床意义。方法采用免疫组化SP法检测40例宫颈鳞癌(癌症组)、20例宫颈上皮内瘤变(CIN组)和15例正常宫颈鳞状上皮(对照组)组织中的pSTAT3、Cyclin D1和VEGF蛋白。结果在癌症组及CIN组,pSTAT3过表达率分别为20.0%、62.5%(P<0.05),Cyclin D1过表达率分别为30.0%、72.5%(P<0.05),VEGF过表达率分别为40.0%、82.5%(P<0.05);对照组均无过表达者。pSTAT3、Cyclin D1蛋白过表达与宫颈鳞癌病理分级、临床分期及淋巴结转移有关(P均<0.05),VEGF蛋白过表达与宫颈鳞癌病理分级、淋巴结转移有关(P均<0.05)。宫颈鳞癌组织中,pSTAT3与Cyclin D1及VEGF蛋白过表达呈正相关(r分别为0.531、0.618,P均<0.05)。结论宫颈鳞癌组织中pSTAT3、Cyclin D1及VEGF蛋白表达增高,三者共同参与了宫颈鳞癌的发生、发展。Objective To observe the expression changes of phosphorylated signal transducer and activator of transcription 3 （pSTAT3）,cyclin D1 and vascular endothelial growth factor （VEGF） in cervical squamous cell carcinoma and to discuss their clinical significance.Methods The expression of pSTAT3,cyclin D1 and VEGF proteins in 40 cases of cervical squamous cell carcinoma （SCC group）,20 cases of cervical intraepithelial neoplasia （CIN group）,and 15 cases of normal cervical squamous epithelium （NE group） was detected respectively by immunohistochemical SP method.Results The overexpression rates of pSTAT3 in the SCC and CIN groups were 20.0％ and 62.5％,the overexpression rates of cyclin D1 were 30.0％ and 72.5％,and VEGF were 40.0％ and 82.5％,respectively （all P ＜ 0.05）.There was no overexpression of all in the NE group.The overexpression of pSTAT3 and cyclin D1 was related to pathological grade,clinical stage and lymph node metastasis （both P ＜ 0.05）.The overexpression of VEGF was related to pathological grade and lymph node metastasis （all P ＜ 0.05）.The expression of pSTAT3 was positively correlated with that of cyclin D1 （r =0.531,P ＜ 0.05） and VEGF （r =0.618,P ＜ 0.05） in the cervical squamous cell carcinoma.Conclusion The expression of pSTAT3,cyclin D1 and VEGF increases in the cervical squamous cell carcinoma tissues,which might contribute to the development and progression of cervical carcinoma.
