摘  要:从传统中药黄精属的两个种中分别提取到两种小分子粗多糖 ( PD,PP) ,用吡啶 -氯磺酸法分别制取到这两种粗多糖的硫酸酯 ( PDS,PPS)。产物经红外、紫外鉴定 ,证实多糖羟基上连上了硫酸基。用离子交换、凝胶色谱提纯得到这两种粗多糖的纯化产物( RPD,RPP)。体外实验研究了这 6种物质对非洲绿猴肾细胞 ( Vero)的细胞毒性和抗单纯疱疹病毒 2型 ( HSV- )的活性。结果表明 ,粗多糖 PP的细胞毒性是由其中的杂蛋白质引起的 ,多糖 RPP无抗病毒活性 ;粗多糖 PD无细胞毒性 ,PD、RPD均有一定的抗病毒活性 ;多糖硫酸酯 PDS,PPS的抗病毒活性与 PD,RPD相比 ,有了很大提高 ,其最低抗病毒有效浓度比 PD,RPD低Two polysaccharides(PD,PP),which all have low molecule weights about 4000,were extracted from Chinese Traditional Medicines.PD has antiviral activity,and PP has cytopathic effect to cells, in vitro .We then use the two polysaccharides to synthesize sulfates(PDS,PPS).The polysaccharides were chemically modified with the method of chlorosulfonic acid pyridine.PDS's antiviral activity,inhibiting the CPE of cells infected with HSV Ⅱ,is raised about 10 times more than PD's.PPS's cytopathic effect to cells is reduced to the same as PDS's,and it has antiviral activity the same as PDS's.On the other hand,the pruity of PP has no cytopathic effect to cells,and the antiviral activity result shows that purity of PP cannot inhibit the CPE of cells infected with HSV Ⅱ.This result shows that the cytopathic effect of PP is from impurity.The PP's content of protein is two times more than PD and it has no other impurity,so we analyze it is the protein that results in the cytopathic effect to cells.
