摘  要:调节原始卵泡形成、起始生长的信号目前仍知之甚少。一个重要的原因就是缺乏一个良好的研究模型。我们以妊娠13d昆明白小鼠胚胎卵巢为研究对象,在5d的贴壁培养后分别用FBS培养液和ITS培养液继续培养共计21d,探讨在不同培养条件下胚胎卵巢卵泡发育和内分泌功能的差异。结果发现在以ITS、BSA和胎球蛋白为血清替代物的无血清培养体系中小鼠胚胎卵巢卵泡发育要优于其在含2%FBS的培养体系中的发育状况。两种培养条件下胚胎卵巢睾酮的分泌水平相当(P>0.05),但在FBS处理组培养液中可以检测到少量雌二醇,而ITS处理组培养液中检测不到雌二醇。结果提示:①妊娠13d的小鼠胚胎卵巢体外培养至一定阶段即具备了合成睾酮的能力,并且在血清中存在的某些未知刺激因素的作用下胚胎卵巢可以获得将睾酮转化成雌二醇的能力;②所建立的以ITS、BSA和胎球蛋白为血清替代物的无血清培养体系更适于小鼠胚胎卵巢卵泡的体外生长、发育。这为日后进一步研究促性腺激素、生长因子、细胞因子等对胚胎卵巢卵泡发育和类固醇激素发生的精确作用提供了一个良好的研究模型。Signals that regulate follicle formation and activation are yet nuclear. One of the most importal reasons is lacking an elegant system for further study. In the present experiments, mouse fetal ovaries in the pregnancy of 13 days were pre-cultured for 5 days, and then they were further cultured in FBS medium or ITS medium respectively for additional 16 days. We found that the fetal ovarian follicular development in ITS group was significantly prior to that in FBS group. The contents of testosterone in cultured medium of both groups were in a similar level (P>0.05). Furthermore a trace of estrodial existed in the cultured medium in FBS group, while there was no detectable estrodial in ITS group. The results show: ①mouse fetal ovaries in the 13-day pregnancy will acquire the ability to secrete testosterone in a cettain cultural period fo vitro. Conversion of testosterone into estrodial does exist in the cultured mouse fetal ovaries with the stimulation of some uncertain factors in sera. ②The serum-free system established in the present experiments is more suitable to culture mouse fetal ovaries la vitro.
