摘  要:为研究内毒素(ET)对大鼠肝Livin基因表达的影响及阳离子A(CA)的保护效应,将72只清洁级SD大鼠随机分为3组:对照组(Ⅰ组)、内毒素组(Ⅱ组)和阳离子A保护组(Ⅲ组)。3组SD大鼠经相应处理后分别在第3、4、8、12小时采集肝样本,采用实时荧光定量RT-PCR和免疫组织化学技术进行分析。结果显示,在mRNA水平和蛋白水平上,ET可下调Livin基因的表达,CA则能明显上调Livin基因的表达。结果表明,这是诱导肝细胞凋亡的途径之一,也是内毒素对肝损伤的重要机制之一;CA对内毒素导致的组织和细胞的损伤有显著的保护效应。The experiment aimed to investigate the effects of endotoxin on expressions of livin gene and the protective effect of cation A(CA) in the liver. Seventy-two SD rats were randomly divided into three groups of control group(group I ), ET treatment group(group Ⅱ ) and Cation A(CA) and ET treatment group(group Ⅲ). The liver samples were taken from SD rats of the three groups at 3rd,4th,8th and 12th post-treatment,and then analysed by SYBR Green I real-time RT-PCR and immunohistochemical method. In result,the change tendency of livin gene was the same at the mRNA and protein levels. ET could downregulate the expression level of livin gene in the liver, whereas CA could up-regulate expression level of livin gene in the liver. The result show that this is one way to induce hepatocyte apoptosis, endotoxin on hepatic injury is one of the important mechanisms and CA has significant protective effects on hepatocyte injuries by ET.
