摘  要:探讨Wnt5a及其受体FZd2在95、108d和122d肌萎缩脊髓侧索硬化症(ALs)转基因鼠大脑内嗅皮层中的表达情况。方法:取95、108d和122dALS转基因鼠大脑皮质,应用免疫荧光和RT-PCR技术,检测Wnt5a和Fzd2在大脑内嗅皮层中的表达变化。结果:免疫荧光结果显示,与同窝野生型鼠比较,95、108d和122d龄ALS转基因鼠大脑内嗅皮层中Wnt5a阳性细胞明显增多。免疫荧光双标结果显示,Wnt5a标记的阳性细胞与GFAP和β-tubulinIII共表达,ALS转基因鼠Wnt5a/GFAP双阳性细胞增多。与同窝野生型鼠比较,ALS转基因鼠Wnt5amRNA水平在95、108d和122d均升高。Wnt5a受体Fzd2表达情况与Wnt5a一致。结论:WntSa和Fzd2在ALS转基因鼠内嗅皮层中表达增多,表明Wnt5a及其受体Fzd2升高与肌萎缩脊髓侧索硬化症密切相关。Objective: To observe the expression changes of Wnt5a and its receptor Fzd2 in the entorhinal cortex of adult amyotrophic lateral sclerosis (ALS) transgenic mice. Methods: The expressions of Wnt5a and Fzd2 in the entorhinal cortex of ALS transgenic mice at the age of 95 d, 108 d and 122 d were detected with immunohistochemistry technique and RT-PCR. Results: Immunofluorescence results showed that WntSa positive cells were increased significantly in the entorhinal cortex of ALS transgenic mice at different ages, compared with wild-type mice. Double-labeling results showed that Wnt5a-labeled cells were co-expressed with GFAP and β-tuhulin III in the ALS transgenic mice and wild-type mice, and WntSa/GFAP double positive cells were increased in ALS mice. Compared with the wild-type mice, Wnt5a mRNA was increased at 95 d, 108 d and 122 d. The expression of Fzd2 was similar to that of WntSa. Conclusion: The expressions of WntSa and Fzd2 are increased in the entorhinal cortex of ALS transgenic mice, which may be associated with the pathogenesis of AIRS.
