摘  要:猪繁殖与呼吸综合征病毒(PRRSV)的感染,不仅造成不理想的先天性免疫反应,还会导致体液与细胞免疫抑制,增加机体对继发感染的易感性,减弱机体的抗病能力。细胞核因子-κB(NF-κB)信号通路在病毒感染与疾病发展过程中发挥重要作用,直接调控宿主先天性免疫反应及细胞增殖、分化和凋亡。研究发现,PRRSV感染早期,某些非结构蛋白(Nsps)会抑制NF-κB信号通路的激活,受NF-κB调控基因如炎性细胞因子、细胞黏附分子、趋化因子及急性反应蛋白等不表达或表达水平下调,从而抑制宿主的先天免疫应答。推测NF-κB在PRRSV感染致病过程中起着关键作用。本研究对NF-κB信号通路激活与功能,NF-κB在PRRSV感染导致的抗体依赖的病毒感染增强作用(ADE)、无力的先天性免疫应答、肺泡巨噬细胞凋亡、肺损伤中发挥的作用及PRRSV Nsps对NF-κB信号通路的影响进行简要阐述。以期为PRRS的防治及疫苗开发提供新的思路。Porcine reproductive and respiratory syndrome virus (PRRSV) infection would lead to suboptimal innate immune responses and a weaker adaptive immune response, thus increasing the susceptibility to secondary infection and weakening the body's resistance to diseases. The signaling pathway of nuclear factorkappaB (NF-кB) plays an important role in the viral infection and diseasees development by regulating host innate immune response and cell proliferation, differentiation and apoptosis. It has been proved that some nonstructural proteins (Nsps) of PRRSV can inhibit the activation of NF-v,B signaling pathway in early infection, host innate immune response would be suppressed when the transcriptional expression of genes regulated by NF-кB did not happen or reduced, such as proinflammatory cytokines, cell adhesion molecules, chemokines and acute reactive protein. All above revealed that the NF-кB could play a key role in PRRSV infection. In this paper, we make a brief summary of the NF-кB signaling pathway activation and function and the role of NF-кB in the antibody-dependent enhancement (ADE), innate immune response, apoptosis of alveolar macrophages, and pulmonary injury PRRSV infection caused, also the non-structural proteins (Nsps) influences on NF-кB signal pathway were expounded. This paper can provide some new ideas for control strategies and vaccine researches of PRRSV.
