摘 要:研究我国艾滋病病毒Ⅰ型 (HIV 1)病毒样颗粒 (VLP)候选疫苗的免疫原性 ,为疫苗进行灵长类实验提供实验依据。将 gag和gag -V3VLP不同剂量 (5 0 μg、10 μg、1μg)在有佐剂 (氢氧化铝 )和无佐剂条件下皮下免疫小鼠 ,然后眼眶采血 ,用ELISA法观察免疫鼠血清中抗体与剂量关系 ,以及中和抗体滴度和抗体IgG亚型IgG1和IgG2a的水平。同时取鼠脾淋巴细胞 ,体外抗原刺激后收取淋巴细胞分泌上清 ,检测细胞因子IFN γ和IL 5水平。结果是gagVLP免疫剂量与是否加佐剂对免疫效果没有显著性差异 (各组P >0 1) ,而 gag -V3VLP免疫鼠剂量与加入佐剂有显著性差异 (各组P <0 0 1)。gag和 gag -V3VLP(5 0 μg ,佐剂 )中和抗体滴度为 1∶2 0和 1∶40 ,同时gag和gag-V3VLP免疫鼠血清 (5 0 μg ,佐剂 )IgG2a和IgG1均有升高 ,IgG2a稍高于IgG1;细胞因子 (5 0 μg ,佐剂 )IFN γ(gag2 40 0 pg/ml,gag -V3 10 0 0 pg/ml)和IL 5 (gag 40 0 pg/ml,gag -V3 2 0 0pg/ml)均有升高 ,IFN γ升高高于IL 5。由此得出 :HIV 1gag和gag -V3VLP候选疫苗能刺激机体产生抗体 ,其抗体有中和活性 ;抗体亚型IgG2a稍高于IgG1,Th1和Th2细胞分泌的细胞因子 (5 0 μg ,佐剂 )IFN γ比对照组升高 16 0倍 ,IL 5比对照组升高 80倍。表明VLP疫苗能诱导机体?This papar is to study the immunogenicity of P55 and chimeric P55 V3 virus like particles (VLP) of HIV 1 subtype B of Chinese strain. C57B1/6 mice were subcutaneously immunized separately with 50μg, 10μg and 1μg of gag and gag-V3 with or without adjuvant (aluminum hydroxide). Antiserum titer was detected by ELISA method. Spleen cells stimulated with correlative antigens were cultured for cytokine assays. The results showed that there was no significant difference in immune effect between different immune doses of gag and the presence or absence of adjuvant (each group P >0.1), but significant difference existed between different immune doses of gag-V3 and the adding of adjuvant (each group P <0.01). When immunized separately with 50μg of gag or gag-V3 with adjuvant, production of IgG2a, IgG1, IFN γ and IL 5 increased, IgG2a production was higher than IgG1, IFN γ was higher than IL 5. The neutralization antibody titers of gag and gag-V3 were 1∶20 and 1∶40 respectively. This study demonstrates that HIV 1 gag and gag-V3 VLP are potent immunogens, they can raise antibody response in mice. The antiserum has neutralizing activity. Simultaneously, gag and gag-V3 VLP producing Th1 type predominant immune response was indicated by IgG2a subclass antibody and IFN γ increasing.
