摘 要:目的:研究探讨玻璃化技术在根瘤菌保藏中的应用,并开展相关实验。方法:分别选取快生根瘤菌、慢生根瘤菌各1株为初始菌株,以30%甘油:15%乙二醇:15%DMSO为玻璃化冻存试剂,对大豆根瘤菌进行玻璃化冻存,分别选取冻存1个月、6个月以及12个月的菌株进行存活率以及固氮酶活性测定,研究玻璃化技术对根瘤菌生物学功能的影响。结果:保藏12个月后,大豆快生根瘤菌、慢生根瘤菌菌株存活率均可达到85%以上,固氮酶活性没有明显下降。结论:所用玻璃化方法及试剂对快生大豆根瘤菌保藏具有很好的作用,对慢生根瘤菌的保藏效果还需进一步实验验证。Objective:The possibility of vitrification technology applied in Rhizobium preservation was studied, and some related experi-ments were done. Method:Sinorhizobiurn fredii and Bradyrhizobium were used as the initial strains, respectively. Vitrification agent was made from 30% glycerol, 15% ethylene glycol and 15% DMSO. Two strains were stored in the vitrification agent for 1 month ,6 months and 12 months, respectively. Then the survival rate and nitrogenase activity of the strains were determined. Result: The survival rate of the strains were more than 85% and the nitrogenase activity did not decrease remarkably after 12 months. Conclusion:There was good effect on Sinorhizobium fredii preservation with the vitrification method and reagent used in this paper, but it needs further study for Bradyrhizobi-um.
