摘 要:目的:探讨止消通脉宁对转化生长因子-β1(TGF-β1)诱导的人肾小管上皮细胞(HK-2)Smad 2、Smad 3、Smad 7的影响。方法:将HK-2细胞用含10%胎牛血清的DMEM/F12(1:1)培养基培养;实验分为6组:空白对照组、单纯TGF-β1诱导组(TGF-β110 ng/mL)、空白血清对照组(TGF-β110 ng/mL+10%空白血清)、中药含药血清低剂量组(TGF-β110 ng/mL+10%低剂量止消通脉宁含药血清)、中药含药血清中剂量组(TGF-β110 ng/mL+10%中剂量止消通脉宁含药血清)、中药含药血清高剂量组(TGF-β110 ng/mL+10%高剂量止消通脉宁含药血清)。药物干预24小时后,荧光定量PCR检测Smad 2、smad 3、Smad 7的mRNA表达。结果:HK-2细胞经TGF-β1诱导后,Smad 2、smad 3的mRNA表达显著上升,Smad 7的mRNA表达下降,与空白对照组相比有显著性差异(P<0.05),但经止消通脉宁含药血清干预后,Smad 2、smad 3的mRNA表达逐步下降,Smad 7的mRNA表达上调,与单纯TGF-β1诱导组相比有显著性差异(P<0.05)。而空白血清无此作用。结论:止消通脉宁能够调控TGF-β1诱导的人肾小管上皮细胞转分化Smad信号通路mRNA表达的作用,在一定程度上具有抑制肾间质纤维化的作用。Objective:To explore the effects of ZhiXiao TongMaiNing on Smad 2, Smad 3 and Smad 7 of hu-man renal tubular epithelial cells (HK-2) which were induced by transforming growth factor (TGF-β1). Method:HK-2 cells were cultured in the media of DMEM/F12 (1:1) containing 10%fetal bovine serum;the cells were divid-ed into six groups:blank control group, TGF-β1 group (TGF-β110 ng/mL), blank serum control group (TGF-β1 10 ng/mL+10%blank serum), low dose group of TCM medicated serum (TGF-β110 ng/mL+10%low dose medicated serum of ZhiXiao TongMaiNing), moderate dose group of TCM medicated serum (TGF-β1 10 ng/mL+10%moderate dose medicated serum of ZhiXiao TongMaiNing) and high dose group of TCM medicated serum (TGF-β1 10 ng/mL+10%high dose medicated serum of ZhiXiao TongMaiNing). After intervened for 24 hours, Smad 2, Smad 3 and Smad 7 mRNA expressions were detected with fluorescence quantitative PCR. Result:Smad2 and Smad3 mR-NA expressions were raised significantly, Smad 7 mRNA expressions were decreased when HK-2 cells were in-duced with TGF-β1, it had significant difference compared with blank control group (P〈0.05), but after intervened with medicated serum of ZhiXiao TongMaiNing, Smad 2 and Smad 3 mRNA expressions were lowered gradually, Smad 7 mRNA expressions were improved, it had statistical meaning compared with TGF-β1 group (P〈0.05). Blank serum presented no effects. Conclusion:ZhiXiao TongMaiNing could regulate mRNA expressions of trans-differenti-ated Smad signal channel of HK-2 cells induced with TGF-β1 and inhibit the fibrosis of renal tubular epithelial cells to a certain extent.
