摘  要:目的探讨家蝇抗菌肽(antibacterial peptides)对幽门螺杆菌(Helicobacter pylori,H.pylori)的抗菌机制。方法通过绘制抑菌曲线、十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(sodium dodecyl sulfate polyacrylamide gel electrophoresis,SDSPAGE)及双向电泳(two-dimensional electrophoresis,2-DE)方法和技术,初步探讨家蝇对幽门螺杆菌作用机理。结果浓度为40μg/mL家蝇幼虫抗菌肽使H.pylori不能达到正常的生长高峰进入对数期,而1d后直接进入衰亡期;通过SDS-PAGE检测发现实验组在分子量约66.4kDa的蛋白条带浓度较对照组降低,而在分子量约40kDa的蛋白条带浓度较对照组增高。双向电泳结果显示家蝇幼虫抗菌肽干扰了H.pylori的抗氧化系统,影响了H.pylori能量代谢和应激作用功能蛋白的表达。结论家蝇幼虫抗菌肽对H.pylori具有明显的抑制作用,是通过抑制其生长、干扰其抗氧化系统,以及影响其能量代谢等方面,发挥家蝇幼虫抗菌肽的抑菌机制。This paper aims to discuss the antibacterial mechanism of antibacterial peptides for Musca domestica on Heli- cobacter pylori. The mechanism for action of Musca domestica on Helicobacter pylori would be discussed preliminarily in this paper by means of the methods and techniques of drawing curves of bacteriostasis, sodium dodecyl sulfate polyacrylamide gel e- lectrophoresis (SDS-PAGE), and two dimensional electrophoresis (2 DE). The antibacterial peptides of Musca domestica larva with the concentration of 40 μg/mL made Hp unable to achieve the normal growth peak and come to the log phase. Instead, it came to the decline phase directly after 1 day. It was detected by SDS-PAGE that, the concentration of protein bands with the molecular weight of about 66.4 kDa of the experimental group decreased compared with the concentration of protein bands of the control group. But the concentration of protein bands with the molecular weight of about 40 kDa of the experimental group increased compared with the concentration of protein bands of the control group. The result of 2-DE showed that the antibacte- rial peptides of Musca domestica larva interfered the antioxidant system of Helicobacter pylori and influenced the expression of functional protein with energy metabolism and stress effect of Helicobacter pylori. It came to a conclusion that the antibacterial peptides of Musca domestica larKa have significant inhibitory effect on Helicobacter pylori. The antibacterial mechanism is brought into play by inhibiting the growth of Helicobacter pylori, disturbing its antioxidant system and influencing its energy metabolism and other aspects.
