摘 要:生物相容性聚合物膜上培养黑素细胞可用于白癜风移植治疗。本文通过甲基丙烯酸酐酰化反应,在壳聚糖中引入双键。以2-羟基-4-(2-羟乙氧基)-2-甲基苯丙酮(I2959)为光引发剂,在365nm紫外光照下进行改性壳聚糖与三乙二醇二甲基丙烯酸酯和聚乙二醇甲基丙烯酸酯的交联共聚,制备改性壳聚糖交联膜(CMMC)。研究表明CMMC具有满足伤口辅料要求的吸水率和水分蒸发透过率,其机械强度优于硫酸钠物理交联的壳聚糖膜,黑素细胞能在CMMC上生长和增殖,并保持生物学活性,满足黑素细胞移植治疗的使用要求。Human melanocytes cultured on biocompatible polymeric membranes can be used for the transplantation treatment of vitiligo. In this study, a modified chitosan having vinyl functional groups was synthesized via an acylation reaction of the chitosan with methacylic anhydride. The copolymerization of the modified chitosan with polyethylene glycol methacrylate and triethylene glycol dimethaerylate was carried out under 365nm UV irradiation with 2-hydroxy-4-(2-hydroxyethoxy) -2-methyl propiophenone (I2959) as an initiator. Crosslinked membranes of the modified chitosan (CMMC) were thus produced. The CMMCs could function as a good wound dressing with required water absorption abilities and water vapor transportation rates, and at the same time possessing mechanical strengths higher than their counterparts physically crosslinked by sodium sulfate. Human melanocytes could grow and proliferate well, and maintain their bioactivities on the CMMCs. The CMMCs meet the requirements for the transplantation treatment of the human melanocytes.
