摘  要:目的探讨丝裂原活化蛋白激酶p38族(p38MAPK)信号通路在鼻炎性嗅觉障碍疾病中的作用,以及p38MAPK特异性抑制剂(SB203580)对嗅黏膜的保护作用。方法选取SD大鼠,随机分成4组,经鼻孔滴入菌液制备鼻炎大鼠模型,静脉注射特异性抑制剂(SB203580)进行干预。HE染色观察大鼠鼻黏膜的损伤情况;免疫组化法检测嗅黏膜磷酸化p38MAPK的表达。结果与空白对照组比较,鼻炎组大鼠嗅黏膜p38MAPK的表达明显增强(P=0.028<0.05);特异性抑制剂干预组的p38MAPK的表达较鼻炎组有所减轻,但差异无统计学意义(P=0.057)。结论 p38MAPK信号通路参与了鼻炎的发生、发展,并导致了嗅黏膜的损害;p38 MAPK特异性抑制剂在炎症过程中可能有减轻嗅黏膜损伤的作用。Objective To evaluate the effects of p38 mitogen activated protein kinase (p38MAPK) on dysosmia of rats with rhinitis and the protective effect of SB203580, the specific inhibitor of p38MAPK, on olfactory mucosa. Methods SD rats were divided randomly into four groups. Rhinitis model of rats was prepared by bacterio-liquid drop and SB203580 was injected intravenously for intervention. Nasal mucosa injury was observed by HE staining; p38MAPK expression was examined by immunohistochemistry method. Results The expression of p38MAPK in olfactory mucosa significantly increased in rhinitis group compared with that in control group (P = 0. 028~0.05). The expression of p38MAPK in olfactory mucosa of specific inhibitor group decreased, but did not differ significantly from that of rhinitis group (P = 0. 057). Conclusion The p38MAPK signal pathway may be involved in the occurrence and development of rhinitis and cause olfactory mucosa injury. The specific inhibitor of p38MAPK may reduce olfactory mucosa injury during the inflammation, which deserves further study.
