摘  要:[目的]研究大花蕙兰茎尖外植体原球茎诱导及增殖培养、褐化污染的影响因素。[方法]以大花蕙兰茎尖作为外植体,研究无菌系建立、诱导培养的灭菌方法与褐化防治、原球茎诱导培养初始培养基的筛选、原球茎增殖培养的影响因素。[结果]结果表明:解决茎尖内生菌污染及褐化严重的问题,两段式灭菌(75%酒精30 s+0.1%升汞10 min+剥去幼叶+0.05%升汞5 min)优于单一式的灭菌;原球茎诱导培养基为1/2MS+2.0 mg/L 6-BA+0.2 mg/L NAA;原球茎增殖培养基为1/2MS+2.0 mg/L 6-BA+0.2 mg/L NAA+2.0 g/L活性炭+10%马铃薯。[结论]该研究可为大花蕙兰的茎尖快速繁殖及工厂化生产提供技术依据。[ Objective] The aim was to study the influencing factors of Cymbidium hybridium protoeorm induction and proliferation culture by u- sing shoot tips, browning prevention. [ Method] In order to develop aseptic system with the shoot tip of Cymbidium hybridium as explants, study on sterilization method of induction culture and browning prevention, the selection of initial media for protoeorm induction, the influencing factor of protoeorm proliferation culture. [ Result] The result showed that two-stage sterilization was better than single sterilization to resolve the problem of endopbytie bacteria pollution and severe browning with the shoot tip. The optimum medium formula for protocorm induction was 1/2MS +2.0 mg/L 6-BA + O. 2 mg/L NAA. The suitable medium formula for protoeorm proliferation was 1/2MS + 2.0 nag/ L 6-BA + 0. 2mg/L NAA + 2.0 g/L AC + 10% potato. [ Conclusion] This research will provide technical references for rapid proliferation and factory-scale production by using shoot tips of Cymbidium hybridium.
