摘  要:目的:探讨麝香保心丸对心力衰竭大鼠肾脏α1-肾上腺素受体(α1-adrenergic receptorα1-AR)和β-肾上腺素受体(β-adrenergic receptorβ-AR)各亚型表达水平的影响。方法:选择雄性Wistar大鼠80只,随机分为正常对照组(10只)、假手术组(10只),其余60只制作心肌梗死后心力衰竭模型,模型成功35只并将其随机分为心力衰竭组(9只)、阳性药物对照组(8只)、麝香保心丸低剂量组(9只)和麝香保心丸高剂量组(9只)。取大鼠肾脏皮质,采用Western Blot分别测定各组大鼠α1-AR和β-AR各亚型蛋白表达水平。结果:假手术组与正常对照组α1A-AR和β1-AR表达水平比较,差异无统计学意义(P>0.05);与假手术组比较,心力衰竭组大鼠α1A-AR、β1-AR表达水平明显下调,差异有统计学意义(P<0.05);与心力衰竭组比较,阳性药物对照组、低剂量组、高剂量组大鼠α1A-AR、β1-AR表达水平明显上调,差异有统计学意义(P<0.05);与低剂量组比较,阳性药物对照组、高剂量组大鼠α1A-AR表达上调更为显著(P<0.05)。各组大鼠α1B-AR、α1D-AR和β2-AR表达水平比较,差异无统计学意义(P>0.05)。结论:麝香保心丸缓解了心力衰竭大鼠肾脏α1-AR和β-AR各亚型表达水平的下调,对恢复肾脏功能、改善心力衰竭起到了有益的作用。同时,与小剂量麝香保心丸相比,高剂量疗效更为显著。Objective:To explore the effect of shexiangbaoxin pills(SXBXP) on expression of renal ctI - adrenergic receptor (Ctl-AR) and B adrenergic receptor (B-AR) subtypes in rats with heart failure. Meth- ods: Eighty male wistar rats were randomly divided into normal control group (n = 10 ) and sham operation group (n = 10). The heart failure model was established successfully in 35 of the rest 60 male wistar rats. The 35 heart failure model rats were randomly divided into four groups: heart failure model group( n = 9) , positive medicine control group( n = 8 ), low dose of SXBXP group( n = 9) and high dose of SXBXP group( n = 9). The protein levels of ctI -AR and B-AR subtypes of renal cortex were measured by western blot in each group. Re- suits: Compared with normal control group, expression of CtlA-AR and B-AR showed no significant difference in sham operation group(P 〉 0.05 ), compared with sham operation group, expression of OtIA-AR and B-AR was decreased significantly in heart failure model group (P 〈 0.05 ). Compared with heart failure model group, expression of cqA-AR and ~x-AR was increased significantly in group positive medicine control group, low dose of SXBXP group and high dose of SXBXP group(P 〈 0.05 ). Compared with low dose of SXBXP group, expres-sion of otlA-AR was increased significantly in group positive medicine control group and high dose of SXBXP group( P 〈 0.05 ). Expression of c^ls-AR,ctlD-AR and ~2-AR showed no difference in each group( P 〉 0.05 ). Conclusion: SXBXP lessen the downregulation of expression of renal oh-AR and 13-AR subtypes in rats with heart failure, and it is also effective to recover renal function and improve heart failure, especially high dose of SXBXP.
