补脑止痫散对戊四氮慢性点燃癫痫大鼠模型GABA及其受体GABAα表达的影响

摘　　要：目的:探讨补脑止痫散对戊四氮(PTZ)慢性点燃癫痫大鼠模型γ-氨基丁酸(GABA)及其受体亚单位α(GABAα)表达的影响。方法:选取60只健康SD大鼠,随机分成6组(空白对照组、癫痫模型组、丙戊酸钠组、补脑止痫散低剂量组、补脑止痫散中剂量组、补脑止痫散高剂量组)。采用连续小剂量腹腔注射戊四氮(PTZ)的方法,建立癫痫大鼠模型(慢性点燃)。获得Ⅲ级以上发作后,停止给药。采用免疫组织化学法,检测各组大鼠海马区GABA及其GABAα的表达。结果:除空白对照组无癫痫样发作外,其余各组都出现Racine分级中的Ⅲ-Ⅴ痫样发作表现。丙戊酸钠组、补脑止痫散高剂量组GABA及GABAα表达明显上调,与模型组比较,有显著性差异(P<0.05)。结论:补脑止痫散对戊四氮慢性点燃癫痫大鼠模型海马区内抑制性递质及受体有明显的上调作用,从而起到保护脑组织、缓解癫痫症状,进而减轻由癫痫导致的脑损伤的作用。Objective：To study the effects of Bunao Zhixian powder on the expression of GABA and GABAα in the brain of Pentylenetetrazole（PTZ）-kindled chronic epileptic rats.Methods：60 SD rats were randomly divided into six groups,control group,epileptic rat model group,Sodium valproate group,low dose of Bunao Zhixian powder therapy group,middle dose of Bunao Zhixian powder therapy group and high dose of Bunao Zhixian powder therapy group.Rat kindling model with epilepsy was established by PTZ-injection.By immunohisto chemistry method detect the protein expression of GABA and GABAα in hippocampus of rats.Results：The occurrence and seizure level were evaluated by using Racine method.The rats with Ⅲ to V seizure level were chosen for the following studies.The expression of GABA and GABAα increased significantly between the Sodium valproate group and the high dose of Bunao Zhixian powder therapy group.There were significant differences by comparing with model group（P ＜ 0.05）.Conclusion：Bunao Zhixian powder plays a significantly increased role in excitatory neurotransmitter and receptor,so as to protect the brain tissue,to alleviate the symptoms of epilepsy and to reduce the role of the damage caused by epilepsy.
