摘 要:目的:分析24个Y染色体短串联重复序列(STR)基因座在江苏汉族群体的基因多态性、基因频率和其他地域人群之间的遗传距离。方法:取2011-2012年来自江苏省的262份无血缘关系的汉族个体,对24个STR位点进行复合扩增.ABI3130自动基因分析仪进行片段分析并进行基因分型。结果:共发现172个STR等位基因,其中DYS385b、DYS449基因座发现14种等位基因,单倍型共发现262个,单倍型多样性为0.9999,统计10个群体遗传距离Rst矩阵,提示江苏人群更偏向于以天津(-0.0018)、河南(-0.00176)、山西(-0.0016)为群体的中原人群,而与湖南(O.01754)和辽宁(0.01464)人群的遗传距离最远(P〈0.05)。结论:获得具有江苏汉族人群特征的遗传学和法医学客观基础数据,对建立具有江苏地区特点的Y—STR数据库有重要意义。Objective:To study the genetic polymorphism,frequencies and genetic distance of 24 Y-chromosomal short tandom repeat (STR) loci in unrelated individuals of Han nationality in Jiangsu province. Methods:Twenty-four STR loci were amplified in DNA samples from 262 unrelated individuals in Jiangsu Han population from 2011 to 2012. All samples of the PCR products were genotyped by ABI 3130 Genetic Analyzer. Results: In total, 172 genotypes and 262 haplotypes were founded, DYS385b, DYS449 loci had 14 genotypes,the diversity of overall heliotypes for the 24 Y-STR loci was 0.999 9. Comparing with 9 other populations,the genetic distance was more closed between Jiangsu and Tianjin (-0.001 8),Henan (-0.001 76),Shanxi (-0.001 6) province, compared to Hunan(0.017 54) and Liaoning(0.014 64) population (P 〈 0.05). Conclusion:We obtained objective basic data in the area of population genetics and forensic medicine,which would be useful for building Y-STR database in Jiangsu Han population.
