摘　　要：目的研究SIVmac251感染对galectin-3诱导恒河猴巨噬细胞凋亡的影响。方法从健康猴和SIVmac251感染猴外周血分离单核细胞,用GM-CSF与IL-6诱导单核细胞分化成巨噬细胞。巨噬细胞与galectin-3共孵育5.5 h后,细胞经PE-annexin V/PerCP-7AAD染色,用流式细胞仪检测细胞凋亡情况。结果无galectin-3诱导条件下健康猴与SIVmac251感染猴巨噬细胞中annexin V阳性细胞的百分率分别为5.08%与4.87%。Galectin-3诱导条件下健康猴巨噬细胞中annexin V阳性细胞的百分率升至17.62%,SIVmac251感染猴巨噬细胞中annexin V阳性细胞的百分率升至45.56%,两者差异具有统计学意义(P<0.01)。结论 SIVmac251感染增强恒河猴巨噬细胞对galectin-3诱导凋亡的敏感性。Objective To study the effect of SIVmac251-infection on galectin-3-induced apoptosis in macrophages of rhesus macaque. Methods Mononuclear cells were isolated from peripheral blood of healthy, or SIVmac251-infected rhesus macaques. The cells were induced to differentiate into macrophages by GM-CSF and IL-6. The macrophages were cultured for 5.5 hours at the presence of galectin-3. The cells were stained with PE-annexin-V/PerCP-7AAD and apoptosis was assayed by flow cytometry. Results The percentages of annexin-V ＋ cells without galectin-3 induction were 5.08% and 4.87% for macrophages from healthy or SIVmac251-infected rhesus macaques, respectively. The percentages of annexin-V ＋ cells with galectin-3 induction was increased to 17.62% and 45.56% , respectively. There were significant differences between the two groups （ P 〈 0. 01 ）. Conclusion SIVmac251-infection enhances the sensitivity to galectin-3-induced apoptosis in rhesus macaque macrophages.
