摘  要:目的探讨脑源性神经营养因子(BDNF)对移植入老年性痴呆(AD)模型鼠基底前脑内神经干细胞(NSCs)的分化的影响。方法取新生SD大鼠海马组织,以无血清培养技术培养获得NSCs并用5-溴-2-脱氧尿苷(BrdU)标记,同时参照包新民等的大鼠脑立体定位图谱制备AD大鼠模型并随机分成BDNF组和对照组,将BDNF和NSCs一起或单独NSCs移植入两组模型鼠基底前脑内,移植后2周取脑切片,行BrdU+神经丝蛋白(NF)、BrdU+胶质纤维酸性蛋白(GFAP)免疫荧光双标染色并且计数比较。结果移植后2周,BDNF组和对照组移植的NSCs在体内分化后都能检测到NF+BrdU和GFAP+BrdU双阳性细胞。BDNF组计数得到NF+BrdU阳性细胞数高于对照组,差异有显著性(P<0.05)。结论 BDNF能促进移植入AD模型鼠基底前脑的NSCs分化为神经元。[ Objective ] To investigate the effects of brain-derived neurotrophic factor (BDNF) on differentiation of neural stem cells (NSCs) transplanted in basal forebrain of Alzheimer's disease (AD) rats. [Methods] The NSCs were obtained from the neonatal SD rat hippocampus by the technique of serum-free culture and were marked with 5-Bromo-2-deoxyuridine (BrdU). The rat model of Alzheimer's disease was induced according to rat stereotaxic atlas of Bao Xin-min, et al. BDNF was used accompanied with the NSCs suspension injected in basal forebrain of AD rats. And rats were sacrificed two weeks after the transplantation. Brain sections were collected for neurofilament (NF)+BrdU or glial fibrillary acidicprotein (GFAP)+BrdU immunofluorescence. The number of the differential cells was compared between the BDNF group and the control group. [ Results ] Two weeks after the transplantation, NF+ BrdU as well as GFAP+BrdU positive cells could be found in the two groups. The number of NF+BrdU positive ceils was increased in BDNF group compared with the control (P 〈0.05). [Conclusions ] BDNF can promote the neuronal differentiation of NSCs engrafted into AD rat's basal forebrain.
