摘  要:目的通过单细胞凝胶电泳(SCGE)和细胞荧光染色技术,研究艰难梭菌毒素A(TcdA)对食管癌干细胞(SP细胞)的杀伤力,探讨其应用价值。方法食管癌干细胞台盼蓝染色检测后,采用单细胞凝胶电泳技术检测TcdA对食管癌干细胞DNA损伤情况;同时通过细胞荧光染色技术研究TcdA作用后的食管癌干细胞发生凋亡的形态学改变。结果 400 ng/mL TcdA作用食管癌干细胞后;细胞DNA被破坏,单细胞凝胶电泳后细胞"彗尾"长度明显增加,荧光强度变大;细胞荧光染色发现实验组的细胞碎裂伴凋亡小体形成。结论 TcdA对食管癌干细胞DNA的损伤及诱发凋亡的特性,可能为食管癌的治疗带来新的希望。[ Objective ] By single cell gel electrophoresis(SCGE) and cell fluorochrome staining, to study the cytotoxicity of esophageal cancer stem cell effected by Clostridium Difficile Toxin A(TcdA), and explore the value of TcdA. [ Methods ] After trypan blue staining, the DNA injury of esophageal cancer stern cell by TcdA will be detected via SCGE, and the apoptosis of cells would be founded through flnorochrome staining. [ Results ] After 24 h, the cell DNA was damaged by 400 ng/mL TcdA, the length of the comet tail was longer and the cell fluorescence intensity was stronger. The cells in the experimental group appeared cytoclasis and apoptotic bodies. [ Conclusions ] TcdA can damage cell DNA induce apoptosis in esophageal cancer stem cell, which will be used to treat esophageal cancer.
