摘  要:骨化三醇,全反式维甲酸和乌索酸是3种能诱导U937细胞单核分化的试剂,但它们对单核分化的影响并不是完全相同。为了研究这3种试剂促进单核分化的机制,采用qRT-PCR来检测它们对一组参与单核分化的转录因子和3个单核细胞表面标记的mRNA表达水平的影响。结果表明:这3种试剂均上调RBSP3,Egr-1,Egr-2和E2F1的表达,此外,ATRA和UA对CD64,CEBPA和E2F3具有相似的影响,但对CD11c和CD18的影响则相反,骨化三醇对所检测的基因除了对CEBPA无显著影响外,对其它基因均上调。以上结果表明,尽管这3种试剂均能诱导U937的单核分化,它们对所检测的基因的影响并不完全相同,这表明它们影响单核分化的机制是不同的。本研究为进一步研究骨化三醇,全反式维甲酸和乌索酸促进单核分化的机制奠定了基础。Calcitriol, all trans retinoic acfd (ATRA) and Ursolic acid (UA) are three reagents that can induce monocytic differentia- tion of U937 cells. However, their effects on monocytic differentiation are not exactly the same. In order to investigate their mechaniams on promotion of monocytic differentiation, the quantitative real time PCR (qRT-PCR) was adopted to detect their effect on the mRNA expression of a group of transcription factors involved in monocytic differentiation and three myelomonocytic surface markers. The results demonstrated that they all upregulate RBSP3, Egr-1, Egr-2 and E2F1. Moreover, ATRA and UA have the similar effect on CD64, CEBPA and E2F3, but have the adverse effect on CDllc and CD18, Calcitriol upregulated all the examined genes except CEBPA~ Taken together, these results demonstrated that although all of the three reagents can promote monocytic differentiation of U937 cells, their effects on the examined genes are not all the same, which indicated that the mechanisms of their regulation on mono- cytic differentiation are different. This study laid a foundation for the further investigation on mechanisms of Calcitriol, ATRA and UA.
