摘  要:建立了一种基于硼酸亲和作用的pH指示剂比色法,并应用于糖和核苷类样品的含量测定。选取苯硼酸为硼酸试剂,以糖类和核苷为测试对象,考察并比较不同波长下采用甲基橙和百里酚兰指示剂时溶液浓度与吸光度变化值之间的线性关系。结果表明,糖类物质浓度在10—300mmol/L范围内,核苷类物质浓度在2~36mmol/L范围内,采用百里酚兰指示剂于600nm下测定,线性关系较好。验证实验结果表明方法具有较好的稳定性,准确度和精密度。A colorimetric method by using pH indicators based on the boronic acid affinity interaction was developed, and was applied to quantitative determination of saccharide or nucleoside. Phenyl boronie acid was selected as the boronic acid reagent. Taking saecharide and nueleoside as the test samples, the linear relationship between the absorbance change value ( logI/Io) and the content of test sample was investigated using the pH indicators (methyl orange and thymol blue) under different wavelengths. The result indicated that the linear relationship of absorbance change value (logI/lo) and contents of saccharide (10 ~ 300 mmol/L) or nucleoside (2 - 36 mmool/L) was good at the wavelength of 600 nm with thymol blue as a pH indicator. Moreover, the stability, precision and accuracy were good.
