摘 要:目的研究利用骨髓库HLA资料建立高匹配诱导多能干细胞(iPS)库的可行性。方法通过对中华骨髓库湖南分库长沙实验室5 236(人)份HLA分型数据统计分析,找出O型HLA-A、-B、-DRB1三座位纯合子数据,计算以纯合子志愿者为假定iPS供者、5 236名随机志愿者为假定患者的匹配几率;同时计算随机志愿者HLA单体型频率,评估以O型HLA高频单体型纯合子志愿者为假设iPS供者的匹配几率。结果在5 236份骨髓供者资料中共发现O型HLA单体型纯合子14种,以此建库可以分别为49.81%(2 608/5 236)、61.29%(3 209/5 236)、79.51%(4 163/5 236)、75.76%(3 967/5 236)及97.78%(5 120/5 236)的假定患者提供5种匹配水平的iPS供者;共检出17种高频率(HF>1%)的单体型,以这些单体型的O型纯合子建库可以分别为64.06%(3 354/5 236)、75.55%(3 956/5 236)、81.11%(4 247/5 236)、78.76%(4 124/5 236)及98.28%(5 146/5 236)的假定患者提供5种匹配水平的iPS供者。结论利用中华骨髓库现有的HLA资料可以建立数量少、效率高的iPS细胞库。Objective To investigate the feasibility of constructing high HLA-matching iPS cells bank using bone mar- row donor' data. Methods A total of 5 236 HLA-A, B, and DRB1 data from Changsha HLA typing lab of Chinese marrow donor program (CMDP) Hunan branch were subjected to estimate the matching potential of the homozygotes with blood group O for random population. Then the haplotype frequencies (HF) were computed and applied to estimate the matching potential of the most frequent haplotype homozygotes with blood group O. Results There dre 14 homozygotes with blood group O. The iPS cells bank constructed using the 14 homozygotes could provide 49.81% (2 608/5 236) ,61.29% (3 209/5 236) ,79. 51% (4 163/5 236) ,75.76% (3 967/5 236)and 97.78% (5 120/5 236) of the patients to find HLA matched donor for 5 match grades, respectively. There are 17 predominant haplotypes (HF 〉 1% ), and the iPS ceils bank constructed using the most frequent haplotype homozygotes with blood group O could provide 64.06% (3 354/5 236) ,75.55% (3 956/5 236), 81.11% (4 247/5 236) ,78.76% (4 124/5 236) and 98.28% (5 146/5 236) of the random population for different match grades respectively. Conclusion It is feasible to construct an efficient iPS cells bank with small scale cell lines using the ex- isting bone marrow donors' data of CMDP.
