摘  要:在优化了流动相组成、比例、流速、洗脱方式和柱温等条件后,选择乙腈和0.5%磷酸水溶液为流动相,流速为1.0mL/min,柱温为35℃,检测波长为217nm,采用梯度洗脱,建立了一个同时分离测定血竭中龙血素A、龙血素B和剑叶龙血素C的高效液相色谱法。该方法在29rain内实现了这3个组分的有效分离与测定,其工作曲线的线性范围对于龙血素A和B均为2.5~250μg/mL,对于剑叶龙血素C为1.0~250μg/mL。将该方法用于测定不同来源血竭样品中龙血素A、B和剑叶龙血素C的含量.相对标准偏荠在5.0%以内.回婀率存95%~110%夕闻.A HPLC method for the simultaneous separation and determination of loureirin A, loureirin B and cochinchinenin C was developed after the parameters affected separation and determination liked the composi- tion and concentration of mobile phase as well as the flow rate, column temperature and the mode for elution were optimized. Loureifin A, loureirin B and cochinchinenin C could be separated and determined effectively within 29 rain by gradient elution with aeetonitrile and 0. 5% phosphoric acid aqueous solution as mobile phase in the flow rate of 1.0 mL/min, column temperature of 35~C and determination wavelength of 217 rim. The linear range for the determination of loureirin A and loureirin B both were 1.25 ~ 250 g/mL, and for the determination of loureirin C was 1.00 -250 g/mL. The satisfactory result for the determination of loureirin A, loureirin B and cochinchine- nin C in dragong blood from natural and artificial inoculation was obtained by the proposed method. The recovery ranged between 95% and 110%. The relative standard deviation (RSD) was below 5.0%.
