摘 要:目的通过在体单向肠灌流实验比较牡荆素及牡荆素鼠李糖苷的肠吸收机制。方法运用大鼠在体单向肠灌流模型并采用HPLC测定肠灌流液中牡荆素及牡荆素鼠李糖苷的含量,计算其有效渗透系数。结果牡荆素及牡荆素鼠李糖苷浓度为80 mg·L-1时,同一时间点同一肠段牡荆素和牡荆素鼠李糖苷之间的有效渗透系数差异无显著性(P>0.05),同一时间点不同肠段(十二指肠、空肠、回肠、结肠)牡荆素和牡荆素鼠李糖苷的有效渗透系数均差异无显著性(P>0.05);牡荆素及牡荆素鼠李糖苷浓度为20、40、80、160 mg·L-1时,同一时间点同一浓度牡荆素和牡荆素鼠李糖苷之间的有效渗透系数差异无显著性(P>0.05),同一时间点不同浓度牡荆素和牡荆素鼠李糖苷的有效渗透系数均差异无显著性(P>0.05)。结论牡荆素及牡荆素鼠李糖苷在肠吸收方面两者不存在明显的差异,且在十二指肠、空肠、回肠和结肠段的吸收均较差,无特殊的吸收窗;不同浓度对牡荆素和牡荆素鼠李糖苷的吸收无明显的影响。Objective To compare the intestinal absorption mechanism of vitexin and rharnnosylvitexin by using in situ single-pass perfu sion method. Methods The in situ rat single-pass intestinal perfusion model was used. The concentration of vitexin and rhamnosylvitexin in the intestine perfusate was determined by HPLC, and the effective permeability coefficinets(Pc,) were calculated. Results As vitexin and rhamnosylvitexin concentration for 80 mg . L^-1 ,the Pelf between vitexin and rhamnosylvitexin in the same time and intestine had no significant difference ( P 〉 0.05 ), and the Peffkin vitexin and rhamnosylvitexin at the same time but different intestne ( duodenum,jejunum, ileu and colon) had no significant difference (P 〉 0.05 ). As vitexin and rhamnosylvitexin concentration for 20,40,80,160 mg . L^-l, the Peffbetween vitexin and rhamnosylvitexin in the same'time and concentration had no significant difference( P 〉 0.05 ), and the Peffin vi texin and rhamnosylvitexin at the same time but different concentration( duodenum,jejunum,lieu and colon) had no significant difference ( P 〉 0.05 ). Conclusions There are no significant differences between vitexin and rhamnosylvitexin in intestinal absorption. There is not a preferential absorption zone in the intestine for vitexin and rhamnosylvitexin, and both of them in the absorption of duodenum,jeju num,ileu and colon were poor. And different concentration of them has no obvious influence for intestine absorption.
