摘 要:基于病例对照研究的试验设计,本试验对β-防御素135基因(β-defensin 135,DEFB135)非编码区和编码区进行单核酸多态性(SNPs)检测,在5’非编码区(5’-UTR)检测到2个SNPs(c.-47 T>A,c.-40C >A),3’-UTR区检测到1个SNP(c.*42C>G),它们处于完全连锁状态.选取c.-40 C >A位点进行基因分型,研究DEFB135基因多态性与家兔非特异性消化道疾病易感性间的相关性.结果发现健康组和肠炎组中等位基因A的频率分别为30%和33%,差异不显著(P>0.05);OR(odds ratio)值为1.11(95%的置信区间:0.849~1.456,P=0.44).低纤维诱导试验中,DEFB135基因在健康组,低轻微组和严重组中的表达量分别为0.0956±0.0273,0.0492±0.0228和0.0475±0.01703,差异均不显著P>0.05.这些结果说明DEFB135基因可能与家兔NSDD不相关.In this case-control study,the polymorphism and expression level β-defensin 135 (DEFB135) were investigated.Primers were designed to amplify untranslated region (5'-UTR,3'-UTR),all two exons.Two single nucleotide polymorphisms (SNPs,c.-47 T > A,c.-40 C > A) in 5'-UTR and one SNP (c.* 42 C > G) in 3'-UTR were detected with 100 % linkage.The c.-40 C > A SNP was randomly selected and genotyped to analyze the association with non-specific digestive disorder (NSDD).The frequencies of A allele in control and case groups were 30 % and 33 % with no significant difference (P > 0.05),respectively.The odds ratio was 1.11 (95 % confidence interval,0.849-1.456,P =0.44).The gene expression level of DEFB135 in three groups were 0.0956 ± 0.0273 (health individuals),0.0492 ± 0.0228 (mild NSDD individuals) and 0.0475 ± 0.01703 (severe NSDD individuals) with no significant difference (P > 0.05).In conclusion,the DEFB135 gene may be not significantly associated with NSDD in rabbit.
